Objective To explore frequencies and polymorphisms distribution of intron 4 of PDCD1 gene, and analysis the susceptibility to systemic lupus erythematosus in indigenous Han Chinese of Anhui Province, and to analyze the influences of many environmental factors on systemic lupus erythematosus.Methods The blood samples of 122 definitive systemic lupus erythematosus patients and 143 controls were collected with case-control study, DNA of the subjects was extracted and amplified by PCR, the SNPs of PDCD1 7809 locus, 7872 locus and 8162 locus were further confirmed by direct DNA sequencing and BLAST software. The relationship of PDCD1 with systemic lupus erythematosus was analyzed by ?test. Environmental factors for SLE were analyzed by univariate and multivariate unconditional logistic regression.Results The allele of PDCD1 7809 locus was G/G among all the subjects investigated,the allele of PDCD1 7872 locus included C/T alleles, 8162 locus included G/A alleles, there were significant difference of the polymorphism distribution of PDCD1 7872 locus and 8162 locus between systemic lupus erythematosus patients and controls (x2= 8.55, x2=11.85, P<0.05). The genotyping frequencies of 7872 locus C/C, C/T and T/T in systemic lupus erythematosus patients were 36.1%, 41.83% and 22.1%, in control were 51.7%, 35.0% and 13.3%, respectively. There was significant difference in the frequency of mutation in 7872 locus between systemic lupus erythematosus patients and controls (x2- 7.411, P<0.05). The genotyping frequencies of PDCD1 8162 locus G/G, G/A and A/A in SLE patients was 50.1%, 20.3% and 28.6%, in control were 57.6%, 20.8% and 22.6%, respectively. There was significant difference in thefrequency of mutation in 8162 locus alleles between systemic lupus erythematosus patients and controls (x2= 7.547, P<0.05). The allele frequency of PDCD1 gene of 7809 locus, 7872 locus and 8162 locus in Anhui Han population were different from other ethical populations in the world, there were different gene locus regulated the PDCD1 gene transcription. Twenty-five environmental factors were found to be related with systemic lupus erythematosus with univariate unconditional logistic regression model. Eight environmental factors were related with systemic lupus erythematosus with multivariate unconditional logistic regression model, mild-salt food, drinking well water and tea were protective factor for SLE, living in cities,history of allergy and touching injurants,taking anti-allergy drugs, noice pollution, were risk factors for SLE. Conclusions These results indicate that the genotype of PDCD1 gene 7809 locus was G,G in indigenous Han Chinese, the SNPs of PDCD1 gene 7872 locus and 8162 locus might affect the susceptibility to SLE. Many environmental risk factors are related to systemic lupus erythematosus, SLE susceptibilities should pay attention to take preventive meatures.
