| Objective To investigate the abnormal expression of FHIT, PTEN, C-myc gene in laryngeal squamous cell carcinom(LSCC) and to learn the mechnism of development and progression of LSCC .Method Immunohistochemical SP metheod was used to detect FHIT, PTEN, C-myc gene protein expression in the paraffin-embedded specimens of 52 cases with LSCC and 23 cases of laryngeal proliferative muscosa and 10 cases of laryngeal normal muscosa. FHIT and C-myc mRNA expression were detected with RT-PCR in 20 cases of fresh laryngeal carcinom samples and its matched contrast groups.Results Positive rate of FHIT, PTEN, C-myc gene protein in LSCC is 46.2%, 48.0%, 76.9%respectively, contrast with proliferative and normal tissues, the difference is significant. FHIT gene protein expression is related to clinical stage and metastasis.C-myc gene protein expression is related to metastasis. RT-PCR show that 14 of 20 samples are deleted in FHIT exon1-4 and 13 of 20 are deleted in FHIT exon5-9. The negtive rate of FHIT mRNA expression is significantly higher than contrast group. 15 of 20 are over expression in C-myc.The positive rate of C-myc mRNA expression issignificantly higher than contrast group.Conclusion FHIT gene expression play a negative role in cell proliferation. The loss of FHIT expression do correlated with the development and progression of laryngeal carcinoma. FHIT may be important candidate supressor gene in LSCC. C-myc gene overexpression is one of the reasons of genesis of LSCC. PTEN gene is possiblly related to genesis of LSCC. |
