The Change Of Platelet CD62P In Different Stages Of Type 2 Diabetes Mellitus Rats And Its Relationship With Renal Disease

Objective To study the change of platelets CD62P in the different stages of diabetes mellitus rats and its significance in the process of diabetic nephropathy.Materials and methods 42 wistar rats were fed on a diet enriched in glucose and fat for 4 weeks to induce insulin resistance(IR) ,then 24 of which were injected intrapentoneally with low dose streptozotocin(30mg.kg-1) to induce type 2 diabetes, the remaining 18 rats fed on the same diet as obesity group ,other 18 wistar rats fed on a normal diet as controls,In the insulin resistance stage, the 12th week after developed diabetes and the 22th week platelets CD62P were determined by flow cytometory(FCM), urinary albumin excretion(UAE) were measured by RIA, and the renal morphologic changes were observed under light and electron microscope .Results In the IR stage .platelets CD62P had a trend to increase but not significantly(6.84*4.13 vs. 4.32*3.38in controls,P>0.05) ,meanwile,urinary albumin excretion(UAE) and renal morphology were not markedly different compared withcontrols .In the 12th week after developed diabetes the expressions of platelets CD62P were significantly enhanced compared with the other two groups (5.97±3.47 in diabetes,0.73± 0.16 in obesity vs.0.81±0.55 in controls,P<9.005),and UAE were not significantly different ,but the glomerular basement membrane had obviously thickened .In the 22th week platelets CD62P were not significantly different among the three groups(2.91±0.86 in diabetes,3.24±1.22 in obesity vs.3.26±0.87 in controis,P>0.05) ,but UAE in diabetic group were significantly higher than the other two groups(13.4±1.2 in diabetes,2.63±0.64 in obesity vs. 3.44±3.18 in controls,P<0.01) and the renal morphologic changes became more seriously.Conelution The expressions of platelets CD62P are in a stasis of changing in the progression of diabetes,Prior to UAE increase .platelets CD62P has significantly increased meanwile, the renal morphology has obviously changed ,So the expression of platelets CD62P may be a more sensitive marker in the diagnosis of early diabetic nephropathy 4 may has a role in the development of diabetic nephropathy .
