To research in vitro chemosensitivity and apoptosis induced by oxaliplatin in human lung adenocarcinoma cell line-A549. We analysised the oxaliplatin inhibition to A549 cells growth by MTT assay, assessed endonucleolysis by agarose gel electronphoresis, observed apoptotic cell by in situ end labeling method and morphological aspects of apoptosis by electron microscope, check apoptosis peak and protein expression of bcl-2 by flow cytometric methods.Results indicated that at dose of 12.5, 25, 50, 100μg/ml, oxaliplatin apparently inhibited growth of A549 cells, the IC50 of oxaliplatin for 24, 36, 48h is 100, 75, 41μg/ml respectively, agarose gel electronphoresis of DNA revealed ladder pattern. In situ end labeling method show apoptotic cells with brown nucleolus. The marked morphological changes included condensed chromatin, nuclear fragmentation and apoptotic body. FCM showed apoptotic peak and tested the down-regulation of bcl-2 at protein level.We drew the conclusion that oxaliplatin might induce apoptosis in A549 cells, the down-regulation of bcl-2 expression may played an important role in apoptosis.
