Purpose1.It is not entirely clear whether povidone iodine and diluted povidone iodine influence the lung cancer cells when being used clinically as a common disinfectants. We demonstrate the tumoricidal effect of PVP-Ⅰagainst lung cancer cells in vitro.2. To evaluate the cytotoxicity effect of PVP-Ⅰ, DW and DDP solution and heating treatment with temperature of 37℃,43℃,or 47℃on lung cancer cells line A549 to select the optimal tumoricidal solutions. We expect to provide the theoretical data for no touch isolation technique of perioperative nursing care.Material and methods1. Research object:Lung cancer cell Line A549, cultivated commonly using RPMI-1640 culture medium containing 10% foetus cattle serum.2. According to the different tumoricidal agent (NS,DW,PVP-Ⅰ,DDP)and the different temperature of heating(37℃,43℃,47℃), the test was devided into 24 groups.Meanwhile NS of 37℃was designed as control group.The apoptosis rate of A549 cell line was measured with MTT assay.3. Morphological observation:Observe the growth state and morphology of cells of each group under inverting microscopes and take pictures before MTT assay.4. Process the data with SPSS 13.0 software package. ANOVA was used to analyze means of multiple groups. Kruskal-Wallis H test and SNK-q was used for the multiple comparison. ANOVA was used for factorial design. Rates were compared with each other by chi-square test. (α=0.05) The results1. The inhibition rate of 0.5% PVP-I for lung cancer cells was 83.64%(37℃),84.71%(43℃),83.62%(47℃)respectively, the effect of PVP-I solutions which have been diluted in 10 times (≥84.75%) is stronger than before(83.64%). Compared with PVP-I and DW solution diluted in 10 times at 37℃, The inhibition rate of the PVP-I solution diluted in 10 times at 37℃increased significantly (P< 0.05).2. Wilconox sign-rank test results showed that there was significant difference in inhibition rates at (37℃,43℃,47℃) among NS. DDP+N sand DDP+DW (P< 0.05). LSD test results showed that there was no significant difference in inhibition rates at (37℃,43℃,47℃) among DW. PVP-I+NS(1:1), PVP-I+NS(1:9). PVP-I+DW(1:9) (P> 0.05).3. Wilconox sign-rank test results showed that PVP-I+NS(1:9) PVP-I+DW(1:9) have the stronger cell killing effect than other groups at 37℃, a significant difference(P< 0.05). ANOVA results showed that DDP+NS and DDP+DW were the most strong tumoricidal solutions at 47℃and there was no significantly difference between DDP+NS and DDP+DW(P> 0.05).6. ANOVA of factorial design results showed that the DDP+DW at 47℃was the strongest tumoricidal agent among all the groups, all the inhibition rates of these groups were less than 100%. Conclusions1. The PVP-I have a very strong inhibition to lung cancer cell, PVP-I diluted in 10 times is stronger than original liquid and distill water at 37℃.2. The effect of DW, PVP-I and PVP-I diluted solution are not temperature-dependent while DDP shows positive relationship with temperature. Thermotherapy (≥43℃) has the strong inhibited effect on lung cancer cells. The inhibition increased when the temperature is up.3.0.5% PVP-I diluted in 10 times has the strongest effect on lung cancer cells at 37℃for 10 min among all tested solutions, which is even 10 times stronger than DDP. Either SN or DW is the ideal thinner for PVP-I, no difference in inhibition.4. The tumoricidal effect of the DW,PVP-I and PVP-I diluted solution are nearly the same at 43℃, which means flexible choices for scrub nurse.5. DDP solution has the strongest effect on cancer cells at 47℃among all tested solution. Comparing with only DDP treatment, significant synergistic effect was observed when combined with the temperature of 47℃for 10 min. Both NS and DW are the ideal thinner for DDP.6. DDP+DW at 47℃is the strongest tumoricidal agent among all the groups. All the tested drugs in the study have the tumoricidal effect for lung cancer cell, but are not able to destroy all the tumor cells, which could not completely eradicate the risk of implantation metastases in surgery.
