Objective To investigate mechanism and effect of Tangke in therapy Diabetic mice Caused by Alloxan.Methods The model of diabetics mice was induced by tail vein injection of Alloxa(25mg/Kg).The mice were divided into 5 groups :normal-control group, model group, Tangke higher dose group , Tangke lower dose group and Yuquanwan group. After two weeks treatment, blood glucose, insulin, glucagon, superoxide dismutase(SOD) , malondialdehyde(MDA) of serum were measured and skeletal muscle GLUT4mRNA was analysed with RT-PCR.Meanwhile, islet cell were observed with microscope and isle B cell expression were measured by immunohistochemistry.Result Blood glucose, glucagon and MDA of Tangke treated group was significantly lower and insulin, SOD of that was significantly higher than those of model group(P<0.05 ,P<0.001). Tangke could ameliorate the situation of islet cell; There was a significant increase in the expression of islet B cell . Skeletal muscle GLUT4mRNA ofTangke treated group was obviously higher than that of model group(P<0.05).Generally, the effect of Tangke higher dose group was better than that of Tangke lower dose group;but,there were not obviously different between Tangke lower dose group and Yuquanwan group.Conclusions The results suggested that Tangke could decrease the blood glucose, partly through increasing SOD to eliminate free radicals and lower the destruction of islet 13 cell, ameliorating islet a cell, increasing insulin in blood and increasing the expression of Skeletal muscle GLUT4mRNA.
