Apoptosis occurs in traumatic brain tissue, and nervous apoptosis may be a kind of modes that neurons died. Nervous apoptosis happens in the animal model with traumatic brain injury. It can be seen in the contusive brain tissue of humans. Many reviews on experimental animal model with nervous systemic injury suggest, apoptosis and anti-apoptosis exist in nervous death simultaneously, and Caspase family plays a very important role.Naloxone can block endogenous opioid receptor, and then stabilize lysosomal membrane, amelioration microcirculation, degrade endothelin and tumor ne rosis factor and so on, so that protect the neurocytes. However there are few literature about anti-apoptotic effect and its mechanism of naloxone in traumatic brain tissue. To investigate anti-apoptotic effect and its mechanism of naloxone in traumatic brain tissue, 77 healthy rabbits were enrolled in a random control study and divided into normal group, control group, high dose group and small dose group, Improved Feeney free dropping objective mode is used to produce severe craniocerebral injury of all rabbits. After formation of models, NS, naloxone injection of high dosage or small dose were administered systemically inrraperitoneally to three groups of rabbitsrespectively in 2 hours, and In our study, 77 healthy male rabbits were euthanized in 15days, histopathology studies of TUNEL and Caspase-3 were made, and the number of apoptotic nervous cells Caspase-3-positive neurocytes were observed and the differences among the values of each group were compared, in order to investigate the anti-apoptotic effect of naloxone to experimental traumatic brain injury and its message-transmitting-mechanism.Materials and methods1.Subject: 77 healthy rabbits were divided randomly into normal group, control group, high dose group and small dose group respectively with 17, 20, 20, 20. There was no significant difference among the average values of body weight in four groups by F test (P=0.6009) .2. Model: Except normal group, in three groups , a bone window was opened in left parietal bone (diameter: 8.0mm). And improved Feeney weight-drop method was used to produce severe craniocerebral injury of each rabbit.3 . Management: Except normal group, in 2 hours after formation of model, in the control group, 2ml NS had been administered systemically intraperitoneally for 10 days, in high dose group, naloxone had been (0.3mg/Kg for first 3 days and 0.1mg/Kg for next 7 days), and in small dose group, naloxone had been (0.01mg/Kg) for 10 days.4. Sampling: 77 rabbits were killed respectively in 15 days after injury. Then paraffin specimen sections were made after all the brain tissue completely taken out.5. Detection: Apoptosis were detected by TUNEL and Caspase-3 was detected by immunohistochemisty.6.Observation: The positive cells of TUNEL and Caspase-3 were counted under light microscope and then the labeling index (LI) were calculated.7 鈥?Medical Statistic Analysis: All data were statistically analyzed with SPSS/11.0 for Windows.They were expressed as mean plus or minus standard deviation (Fix) and 95% CI. Hypothesis testing methods used included reliability analysis, F test and x2 test. In the statistical analysis of this study, the level of hypothesis testing( a ) was P<0.05 in order to avoid false positives( a -error).Results1 .Basic Information(1) Comparison Between the Values of Death Rate in Three GroupsIn the experiment, except normal group, a few of rabbits in the other groups have died from infective diarrhea. Death rate of rabbits was 20.0% in the control group, 15.0% in the high dose group and 25.0% hi the small group. There was no significant difference between death rate hi three groups by x test (P=0.7296).(2) Comparison Between the Average Values of Body Weight in Four GroupsExcept the dead rabbits, the average values of body weight is (1.98 0.166) in the normal group, (1.86 0.156)Kg in the control group, (1.94 0.225)Kg in the highdose group and (1.99 0.322)Kg in the small dose group.
