Objective: To investigate the expression of connective tissue growth factor (CTGF) in renal tissues of patients with IgA nephropathy in Guangxi and its relationship with pathology manifestations of kidney. Methods: According to the classification criterion of IgA nephropathy by WHO in 1982 by which IgA nephropathy was divided into 5 grades, 56 natives of GuangXi with IgA nephropathy were classified into four groups, with 8 in grade 1 group, 13 in grade 2 group, 21 in grade 3 group and 14 in grade 4 and 5 group. The expression of CTGF, transforming growth factor- β1(TGF- β1), fibronectin(FN),and collagen IV(COL-IV) in four groups was detect by Immunohistochemical staining and the relation between the expression of CTGF and serum creatinine (Scr) and creatinine clearance rate (Ccr) was studied. Results: CTGF was mainly produced by tubule endothelial cells in renal tissue, and the expression of CTGF was positively correlated with pathology manifestations of kidney (P < 0.01). Futhermore, the expression of CTGF was also positively correlated with expression of TGF-β1, FN and COL-IV in renal tissue and Scr and negatively correlated with Ccr (P=0.000).There was no expression of CTGF in glomeruli and vessels. Conculsion: 1. CTGF is mainly expressed by renal tubule and increased significantly with the grade of IgA nephropathy being higher. It may play an important role in the development and progression of renal fibrosis. 2. CTGF and TGF- β1 weredetected not completely in the same kinds of cells. CTGF was expressed mainly by tubule endothelial cell, but TGF- P i was detected in both glomeruli and the surrounding of vessels. 3 With renal lesions getting severer , the expression of FN and COL-IV increase significantly and is positively correlated with the expression of CTGF. 4 the expression of CTGF is positively correlated with Scr and negatively correlated with Ccr (P < 0.01), which indicates that CTGF is associated with the decrease of renal function.
