| Granule membrane protein 140( GMP - 140) is a granule membrane glyco-protein which exists on the a - granule membrane of quiescent platelete , and expressed on the surface of platelete membrane when the platelet is activated. It is a marker of activated platelete.GMP - 140 caused the adhesion and connection of platelet - neutrophil, platelet - endothelium and platelet - fibiinogen, then a good deal of platelets and fibrinogen aggregate together. It will accelerate the formation of arterial thrombus. After the occurance of arterial thrombosis, the platelet is activated un-reversible. GMP - 140 is then released continually. Many studies indicate that GMP -140 in acute cerebral infarction (CI) is increased significantly compared with the control group and is positivly correlated to infarction volum, which is indicated that GMP - 140 share the brain injury in the early period, and degenerate the disease.There is research found that cigarette smoking can increase the GMP -140 level, and it is positivly correlated to somking index (SI). It is said that platelet is activated in smokers.As for smokers, the platelet is activated through the endothelium injury and microvascular pathological change, which is accelerate the atherosxlerosis ( AS) and cause the cerebral infarction.Our study use the Flow Cytometer(FCM) for testing GMP -140 ,so as to explore the function of smoking in atherosxlerosis and cerebral infarction.Sbjucts and MethodsThe subjucts were devided into four groups, that is smoking infarctiongroup, nosmoking infarction group, smoking healthy group and nosmoking healthy group. There are 30 subjucts in every group. All of them are 60 - 65years old people without chronic disease,hyerlipidemia,and with normal weight index. All the patients come from our hospital. They all suffer from their first brain infarction. The blood sample was taken within serven days after their morbidity. Healthy group subjucts are people who come here to take their examination.The subjucts are taken samples in the morning after their overnight fast. The first 1. 0ml vein blood is abondoned. Take another 1. 0ml vein blood into a plastic tube with 106mmol/L EDTA and mix them evenly. Place the anticoagulant vein blood 5 ul and CD62pPE 10 ul in another plastic tube and mix them carefully. The tube is free from light for 20 minutes. Add 2 ~ 8C 1 % polysorbate 1.0ml and mix them again. Place it into a 2 ~ 8C refrigertor. Test the platelet GMP -140 on the Flow Cytometer after 2 hours and within 24 hours.Results1.
