Essential hypertension (EHT) is a kind of common cardiovascular disease. With the rising of peopled living condition and the elders aging, its become a severe public health problem which has effected people's living quality. There are many kinds of causes for EHT, but the precise mechanism is not elucidated till now, so it has become the hot point of researching. Chlamydia pneumonia (C. p) , as the third species of Chlamydia genus, is an obligate intracellular pathogen. Chlamydia pneumonia is a common human pathogen which can cause respiratory tract infection such as pneumonia, pharyngitis and sinusitis. Recently, several studies have demonstrated an association between C. p infection and cardiovascular diseases or cerebrovascular diseases, but the results are still in argued. In order to clarify the precise relationship between the C. p infection and the EHT, we investigated the epidemiological markers which concerned with EHT and measured serum immunoglobulin A and G (IgA, IgG) antibody to C. P.Subjects and Methods1. Subjects: 56 patients with EHT were selected and at the same time 35 healthy controls were randomly selected in Beipiao city, Liaoning province. All the subjects who had acute upper and lower respiratory tract infection, acute bronchitis or other lung infections were excluded from this study.2. The diagnosis standard of EHT and the methods of measurement: According to 1999 WHO - ISH guidelines for the management of hypertension, hypertension was defined as a systolic blood pressure (SBP) 5=140mmHg and/or a diastolic blood pressure ( DBP) 90mmHg. The measurement of blood pres-sure should be carried out according to the unified standard under standard conditions. The details of measurements as follow: (l)the patients should relax at least 5 minutes before the test and should not smoking and drinking coffee within 30 minutes. (2) the patients should sit in the chairs, naked the right upper arm, let the elbow and the heart at the same level. (2)the systolic blood pressure is the I stage of K" sound and the diastolic blood pressure is the V stage of K' sound (the vanish sound). (4) repeated test after 2minutes and using the mean value of these two tests (if the difference of the measurements over 5 mmHg, test again after 2 minutes and then using the mean value of these 3 tests).3. The contents of epidemiological survey: the questionnaires were used by the way of inquiring and measurement in sites. The contents of survey included: general characteristics, family history of hypertension, alcohol intake, smoking habits and so on. Five milliliters blood samples were drawn from an antecubital vein after an overnight fast. After centrifugation, the serum fraction was removed and frozen in aliquots at -70 until assayed.4. The steps and the methods of detecting indicators.-4. 1 The presence of specific - IgA and IgG antibody to C. p was assayed using the solid -phase enzyme immunoassay (E1A) kit commercially available from Labsystems company ( Labsystems, Helsinki, Finland) . The concrete procedures ; using sample diluent of reagent box diluted the serum specimens according to the rate of 1:101; CD Reserve 2 empty wells for the blank and 3 for borderline control, positive control and negative control respectively; and others pipette 10l of the diluted specimens ( 1:101) into Microstrip; incubate for 1 hour at +37 ; empty the wells into a suitable biohazard container or aspirate the well contents with a washer; (2) Pipette 100l of enzyme conjugate, incubate for 1 hour at + 37 , and wish the Microstrip with washer for 5 times; (3) Pipette 100l of the TMB - substrate solution into each well, incubate for 30 minutes at room temperature in a dark place; (4) Stop the enzyme - substrate reaction by adding 100l of stopping solution (0.45 M H2S04) into each well and then measure the absorbance immediately at 450nm ( Bio - rad Company 550 ) ; Using the formula to calculate the EIU value of IgA antibody; EIUspl = [ ( Aspl -Ablank)/( Acal - Ablank) ] x30.
