Objective We aimed to observe the changes of the VEGF protein and VEGFmRNA expression in the retinea of diabetic rats which were treated by the angiotensin II receptor I antagonist Valsartan,to reveal the therapeutical mechanism of suppressing the rennin-angitensin system in the diabetic retinopathy. Method 60 rats were divided into normal ,diabetes mellitus and treatment group randomly.The rats model of experimental diabetes mellitus were induced by the peritoneal injection of STZ,and then were treated with Valsartan added to the water. Used immunohis-tochemistry and in situ hybridization to test the expression of VEGF, c-jun protein and VEGFmRNA in retinal paraffin slide during different periods and in different groups of the rats. Results (1)In normal group,1 month dibetes mellitus and 1 month treatmen group,there was no expression of VEGFmRNA and VEGF protein in the retinal tissues.In 3 month diabete mellitus group ,the expression of VEGFmRNA and VEGF protein was positive. The VEGF protein expression mainly located in cells of ganglionic layer and vessels, The VEGFmRNA expression mainly located in cells of ganglionic layer and inner nuclear layer,but not in vessels.In 6 month diabete mellitus group, the expression of VEGFmRNA and VEGF protein was srong positive,and the location is similar to the 3 month diabetes mellitus.In 3 and 6 month treatment groups,the expression of VEGFmRNA was weak positive and positive respectively,similar to the VEGF protein .And their location was also similar to the diabetes mellitus group. (2)As to c-jun,there was no expression in normal group,positive expression in 1 month diabete mellitus group and strong positive expression in 3 and 6 month diabete mellitus group .However ,there was only weak expression in each treatment group. Conclusion (1)The expression of VEGFmRNA and VEGF protein significantly increased in the retinal tissue of diabetic rats,and the angiotensin II receptor I antagonist Valsartan strongly inhibited their expression; (2)C-jun was a important introcelluar component regulating the ecpression of VEGF, and c-jun was regulated by AngII; (3)Valsartan can be used to prevent and control the early diabetic retinopathy.Postgraduate Shao Leping (Ophalmology)Directed by Prof. Niu Yingjun...
