objective: To investigate relationship between ACE, angiotensin II type I receptor gene polymorphism and the effect of antihypertension reatment byValsartanMethods: fifty-five hypertensive subject with age between aged 42 and 65 years consent to take apart in this study . absence of secondary forms of hypertension, subjects with a history of cardiac or hepatic, diabetes mellitus were excluded ,no symptoms of heart failure, stroke and lower-linber arterial disease All patients have received Valsartan (80mg/qd) for 8 weeks and no antihypertensive therapy for preceding two weeks , all patients' blood pressure were measured at 2,4,6,8week in the clinic. DNA was extracted from peripheral blood leucocytes.The insertion/deletion polymorphism at the intron 16 of ACE gene And A-C polymorphic site at 1166 nucleotide position in the 3 untranslated region of AT-R were determined using restriction length polymorphism analysis of DNA generate by polymerase chain reaction (PCR-RFLP). Results: The patients' number is 4 of I I genotype ,added to the genotype ID â‘ The frequency of D allele is 59.10%, and the frequency of I allele is 40.90%. â‘¡ The frequency of A allele is 90.9%, and the frequency of C allele is 9.1%. ACE LociBlood pressure was significantly reduced in both groups, there is a remarkable difference of reduce in SBP and DBP between group ID and group DD on the 2w and 4w(p<0.05),but not different significantly after 6w. The efficiency between DD and ID group weren't different significantly (p>0.05).ATR Locithere is no difference in 2,4.6.8 weeks in two group .The efficiency between AA and AC group weren't different significantly (p>0.05).Conclusions: â‘  The result indicate that patients with I allele showed a more quickly respond to Valsartan therapy than patients with D allele.. While this difference diminish gradually with the treatment time came longer.â‘¡ There was no difference between AA and AC genotype in blood pressure reduce and efficiency.
