Research Of MRNA's Expression And Relationship Of Vascular Endothelial Growth Factor And Matrix Metalloproteinases-7 In Colorectal Cancer

Objective: To examine the expression of VEGFmRNA and MMP-7mRNA in colorectal carcinoma and to clarify the relationship between the mRNA expression and the clinicopatholoic features in patients with colorectal carcinoma.In order to clarify the relation between VEGFmRNA and MMP-7 mRNA in the carcinoma metastasis. Materials and Methods : All tissues sampales were taken from 56 patients with colorectal cancer after surgery in the Surgery Department of Taian City Central Hospital from May ,2002to May ,2003 .Each case includes cancer tissue and the tissue beside cancer tissue(from the cut line 5cm) .All sampals were carried to experement department in 30 minutes to store for use in -80 C refrigerator . The pathological diagnosis was performed to described the degree of differentiation and invasion .The clinicopathologic stage was classified by the standard of Dukes .All of the speciments were determined for mRNA expression of VEGF and MMP-7 by using RT-PCR technique. Results: The VEGFmRNA positive expression rate in colorectal carcinoma was 51.8%and it almost didn't expresse in normal tissue.(p<0.05).VEGF mRNA expression was related with the degree of differentiation ,lymph node metastasis ,liver metastasis and clinical staging (p<0.05).MMP-7mRNA have expressed in 54 patients with colorectal carcinoma. MMP-7 mRNA was not related with cancer celldifferentiated degree(p>0.05),but related with the clinical staging and several clinicopathological factors including lymph node and liver metastasis (p<0.05).The MMP-7 mRNA quantity in the VEGF mRNA positive expressive group was significantly higer than the negative group.(p<0.01) Conclusion: The expression of VEGF mRNA is associated with the MMP-7 mRNA expression quantity.They are positively correlated with clinical staging and lymphtive and livermetastasis of colorectal cancer tissues.The examnation of VEGFmRNA and MMP-7mRNA may be a valuable marker for the diagnosis and prognosis of colorectal carcinoma and provide a method for clinical biological treatment.
