Purpose: Rectal Carcinoma was one of the highest incidence malignant tumor in our country, and degree of malignant was very high, which was easy to micrometastasis by lymph and blood vascular in the early stage, although is treated by operation and radiation, drug therapy, there is rather high ratio of recurrence. Recently it has been reported that the tumor tiny vascular creation and degradation of the extracellular matrix were involved in the invasion and metastasis. Vascular Endothelial Growth factor (VEGF) and Matrix metalloproteinases (MMPs) were present hotspot of gene level research of tumor invasion and metastasis. The gene level expression of VEGF and MMP-7 in the rectal carcinoma was researched in the paper in order to discuss rectal carcinoma microvascular creation, invasion And metastasis mechanism and the action in tumor invasion and metastasis between VEGF and MMP-7, elementarily discuss the correlation of the gene related the rectal carcinoma metastasis , clinical stage , invasion and Metastasis. We may process forward judgment for rectal carcinoma biology characteristic and forehand stage, which was the basis for judging the degree of alignment, treatment and prognosis,which reduced the opportunity of recurrence and metastasis.Methods: The thirty cases of rectal carcinoma tumor tissues which were removed by operation contrast with normal mucosa whose distance was above 10 centimeter from tumor, each case tissue simultaneity amplified -actin as internal contrast, using Dukes stage as basis, carrying out reverse transcript-polymerase chain reaction (RT-PCR) technology , electrophoresis belt images were attained by amplified products in gelatum imaging system. We may process quantity analyses and statistic.Results: l.The 26cases(87%) of the 30 cases rectal carcinoma amplified VEGFcDNA Namely expression positive, and 15cases(50%) VEGF expressed in normal mucosa. Among 13 cases with positive expression of tumor tissue and normal mucosa, the expression level of VEGF in cancer tissue was significantly higher than that in normal mucosa; the 29 cases (96.7%) of 30 cases rectal carcinoma amplified MMP-7cDNA namely MMP-7 expression Positive, only 1 case (3.3%) among normal mucosa feebly positive expression,there was statistical difference. The results show that the expression of VEGF and MMP-7 gene enhanced in rectal carcinoma, and VEGF and MMP-7 may be as the biology index of detecting rectal carcinoma.2.The positive expression of VEGF and MMP-7 in the cancer tissue correlates closely with clinical rectal carcinoma Dukes stage. Except DukesA and DukesB expression level no statistical difference, there was significantly difference among each other stage. Along with stage gradually development, the positive expression level of VEGF and MMP-7 in the cancer tissue gradually increased, and the positive expression of VEGF and MMP-9 in cancer tissue was not related to sex, age, the size of the tumor, tumor tissue differentiation. Indicating the VEGF and MMP-7gene expression play an important role in the creation, invasion and metastasis of rectal carcinoma.3. The positive expression of 26 cases VEGF in the tumor tissue has MMP-7 expression positive, and MMp-7 negative expression has no VEGF positive expression, but positive expression Ratio of VEGF and MMP-7 simultaneity was 87%.
