To investigate the anti-inflammatory and immunodulatory actions of total flavonoids of saciente (TFS) and its mechanism, the acute, chronic inflammatory and immune inflammatory models (adjuvant arthritis, AA) were established, meanwhile a series of immumopharmacological experiments in vivo and in vitro were adopted. The main results as follows:1. Anti-inflammatory actions of TFSTFS(120, 240, 480 mg-kg-1, ig) could significantly inhibit the mice ear swelling, the rat palm swelling and the rat cotton pellet granuloma induced by dimethylbenzene, carregeenin, and cotton pellet wool respectively. The primary inflammatory reaction in adjuvant arthritis (AA) induced by Freund's complete adjuvant (FCA) was also inhibited by TFS. The results showed TFS have a remarkable anti-inflammatory action.2. Immunoregulatory actions of TFSTFS(240 , 480 mg-kg-1, ig) apparently improved carbonic clearances of macrophages (M ) and the hemolysin level in serum and splencyte in mice immunosuppressed by Cy . Delayed-type hypersensitivity (DTH), which suppressed by Cy, was enhanced by TFS (120, 240, 480 mg-kg-1, ig) in mice. These results suggested TFS could recover nonspecific, humoral and cellular immune function in mice with immunosuppression. TFS (240 mg-kg-1, ig) significantly increased both CD4+and CD8+ cell, especially influenced on CD4+ cell, which rendered the ascending ratio of CD4+/ CD8+ cell. The results suggested the immunoregulatory actions of TFS might be related to regulating T cell subtype.TFS(10-8~10-4mg/L) not only increased ConA and LPS induced splenocytesproliferation and IL-2 production of splenocytes, but also reduced the elevated IL-1 production and PGEi release from peritoned macrophate (PM ) in AA rats in vitro. 3. Therapeutic actions and its mechanism of TFS on AA ratsTFS (80, 160, 320mg-kg-1, ig) could significantly inhibit secondary inflammatory reaction(secondary swelling , multiple arthritis , pathologic change of ankle arthritis) in AA rats.The results in vivo showed that the low response of splenocytes to ConA and LPS and the decreased IL-2 synthesis were restored in AA rats treated with TFS (160, 320 mg-kg-1, ig), while the elevated IL-1 and PGE2 released from PM were also reduced. The results suggested the therapeutic action of TFS on AA rats be related to its immunoregulatory actions.
