To observe the anti-inflammatory and immunomodulatory effect of total flavonoids extracted from Chrysanthemum indicum (TFC) , and explore the mechanism. The acute, chronic inflammatory and immune inflammatory (adjuvant arthritis, AA) models were established. The levels of cytokine production from peritoneal macrophages including NO, SOD, MDA in AA rats were observed. The main results were as follows:1. Anti-inflammatory actions of TFCThe inflammatory function of TFC was tested by four inflammatory models: dimethylbenzene-induced mouse ear edema, carrageenan-induced rat hind paws swelling, cotton-induced mouse granuloma and FCA-induced AA models. TFC could remarkably inhibit the ear edema induced by dimethylbenzene in mice at the dose of 125,250,500mg.kg-1, the swelling of rats hind paws induced by carrageenan and primary inflammatory reaction in AA rats at the dose of 84,168,336 mg.kg-1 and reduce the weight of granuloma induced by cotton in rats at the dose of 84,168,336mg.kg-1, respectively. The results showed TFC have a significant anti-inflammatory action. Moreover,TFC (500mg.kg-1 ) ig. significantly prolonged the latency of analgesia in hot-plate and tail-flick models in mice.The results showed that TFC have analgesic effect.2. Immunoregulatory actions of TFCTFC(0.005, 0.05, 0.5, 5, 50 mg.L-1) apparently increased the depressed splenocytes proliferation in immunodepressed mice, while it have no effect on splenocytes proliferation in normal mice.
