Objective To study the expression of E-cadherin and matrix metalloproteinase-9 in the progression of cholangiocarcinoma, and the relationship between the expression of them and invasion, metastasis and prognosis. Methods The expression of E-cd and MMP9 in 52 cases of cholangiocarcinoma were examined with S-P immunohistochemical technique. Results The positive rate of E-cd expression in cholangiocarcinoma, tumor-adjacent and normal epithelium of bile duct was 36.5% (19/52), 96%(30/31) and 100%( 10/10), respectively(P<0.01); The positive rate of MMP-9 expression in cholangiocarcinoma, tumor-adjacent and normal epithelium of bile duct was 84.6%(44/52), 22.6%(7/31) and 0%(10/10), respectively(P<0.01). 2. In 52 cases of cholangiocarcinoma, the significant correlation was observed between low E-cadherin expression and pathological grade, TNM stage, invasion, metastasis and the methods of operation(P<0.05), and no difference could be found between E-cd expression and sex, age of patients, site and size of tumor(P>0.05); The significant correlation was observed between high MMP-9 expression and pathological grade, TNM stage, invasion and metastasis and the method of operation(P<0.05), and no difference could be found between MMP.9 expression and sex, age of patients, site and size of tumor(P>0.05). 3. In cholangiocarcinoma, E-cd expression was negative correlation with MMP.9 expression. The positive rate of E-cd expression was 25%( 11/44) in 44 cases of positive MMP-9 expression; The positive rate of E-cd expression was 100%(8/8) in 8 cases of negative MMP-9 expression. 4. The one-year's survival rate of patients with positive E-cd expression was significantly higher than that of patients with low ornegative expression(P<0.05); The one-year's survival rate of patients with positive MMP.9 expression was significantly lower than that of patients with weak or negative expression(P<0.05); Multivariate analysis by Cox's proportional hazrads model showed that the expression of E-cd and MMP-9 is an independent prognostic factor of patients with cholangiocarcinoma. CONCLUSION The expression of E-cd and MMP-9 may be linked with progression and invasion, metastasis. Determination of E-cd and MMP-9 will help in predicting tumor metastatic potential and prognosis of patients with cholangiocarcinoma.
