Glioma is one kind of common maglinant tumor in central nervous system,and it is account for 43%-50% in central nervous system maglinant tumor. It is difficult to eradicate by operation. It is not sensitive to approach of accessory treatment used in clinic at present, for example radiation treatment and chemical treatment. Apoptosis is a kind of cell death ways .It is a initiative process in cell death. It is related to tumor happening. Tumor take place because cell original gene activation and prohibitive gene unactivation. If cell apoptosis is controlled, cell will live longer, death ratio will fall and the number of cell will increase. Someone has found that people can treat tumor by inducing cell apoptosis. Tumor happening is related to cell apoptosis. In tumor tissue, apoptosis is recovery mechanism of body resisting cell malignant. Because of cell abnormally growth, tumor taken place. With the studying of apoptosis phenomenon, people pay attention to the treatment of tumor in cell apoptosis aspects. Nucleic acid vaccination is a new treatment to cerebral gliomas. Exterior gene enter cell DNA, then exterior gene combine with cell DNA. Recompose DNA enter animal body. Exterior gene produce antigen in animal body, and immunity system is activated. It can produce immunity answer. It can help eliminating tumor cell. This experiment objective is to explore the antiblastic effect of pvVP3 gene nucleic acid vaccination and pvHN gene nucleic acid vaccination to C6 glioma of rat. This experiment is based on some theories. First, apoptin is product of CAV VP3 gene. VP3 gene can choose inducing way to make cell apoptosis. When p53 gene mutating, it can not induce cell apoptosis. But VP3 gene do not use p53 method. Apoptin do not kill normal cell and have not poisoning effect. Second, NDV HN gene do not come into being noxious effect. It can increase body immunity effect. This experiment adopt following methods. After having successfully cultivated C6 glioma cells and had an inoculation in Wistar rats, the author set up the animal models of Wistar rats bearing C6 glioma. The author assigned stochastically the rats into three groups and gave each group different treatment respectively. Fore-and-after weight difference and tumor average diameter, the ultrastructural changes were detected, so was the ratio of apoptosis. After completed experiment,I achieve these results: 1,After successfully cultivated C6 glioma cells, Wistar rats behave slowly and some rats have poignant ache.Some rats splenetic. 2,pvHN group and pvVP3 group tumor growth speed are slower than control group. PvVP3 group tumor growth speed is lowest. Fore-and-after weight difference and tumor average diameter in pvHN group and pvVP3 group are little than control group (P<0.05). Fore-and-after weight difference and tumor average diameter in pvVP3 group are the least (P<0.01). After treatment, there is a rat whose tumor disappeared in pvVP3 group, and there is a rat whose tumor average diameter is 35mm in pvHN group. 3,The ratio of apoptosis in pvHN group and pvVP3 group was (33.73%±2.15%) and (68.46%±2.47%) respectively.The ratio of apoptosis in control group is (12.87%±1.37%). Then, we can achieve some conclusions: 1, pvVP3 gene nucleic acid (GNA) have remarkable restraining effect to C6 glioma. Because pvVP3 gene nucleic acid (GNA) can induce cell apoptosis, we can achieve this result. 2, pvHN gene nucleic acid (GNA) have remarkable restraining effect to C6 glioma. Because pvHN gene nucleic acid (GNA) can induce cell apoptosis, we can achieve this result. 3, pvVP3 gene nucleic acid (GNA) have stronger inducing cell apoptosis effect than pvHN gene nucleic acid (GNA). 4, Typical changes of apoptosis were detected in ultrastructural observation in pvHN group and pvVP3 group. 5, The ratio of apoptosis in pvVP3 group is higher than in pvHN group.
