| Alcoholism is one of important factors in cerebrovascular diseases. Nowadays many studies show that the mechanism which contribute to cerebrovascular diseases and cerebric lesion after alcoholism includes hyperlipemia, hypertension, B-endorphin, high blood viscosity, arrhythmia, volume of blood flow reducing in local brain and ion derangement. But there are no reports about if alcoholism may bring about pathologic changes of cerebrovascular self and bring about blood vessel rearrangement, so as to result in cerebral blood supply reducing, thus go step further lead to idiophrenic degeneration and lesion. Therefore this thesis depended on anmimal test model to observe cerebrovascular pathologic changes and cerebric lesion after alcoholism and provide objective basises, so that we can adopt related protective measures to effectively reduce cerebrovascular diseases for those people who drink a lot for a long time after alcoholism in advance.[Objective] This study depended on alcoholic animal test model with Wistar rats to observe the pathologic changes of cerebrovascular self and related cerebric lesion after alcoholism by microscope and electron microscope. It would provide objective basises to reduce cerebrovascular diseases for those people who drink a lot for a long time.. [Method] Wistar white rats are devided into two groups at random. There are 22 rats in experimental group of alcoholism and 16 in control group. Alcoholic group: every rat was fed with 50% alcohol two times everyday according to 8ml/Kg,the interval of time is 6 hours. Two weeks later, every rat was fed according to 10ml/Kg.A week later they were fed according to 12ml/Kg,the intervals of time are all 6 hours. The total time of this test is four weeks. Control group : every rat was fed with the same dosage of 0.9% sodium chloride at the same time, and the total experimental time is four weeks, too. During the experiment we observe the two group of rats with their general condition, measure their weight periodically. After four weeks of test time, the brains of two groups of rats are obtained after perfusion fixation with fixation fluid ascending aorta. Then refixed preparation, dehydration, embedding, section, staining, observation with microscope and electron microscope. [Result] Rats in alcoholic group present trichoxerosis, trichomadesis, malnutrition, loose stool and so on. Some rats present symptoms of cerebral apoplexy in addition, such as acroparalysis and Horner's syndrome. Three weeks later the difference of weitht of rats of the two group show statistical meaning (p<0.05).There are changes in histomorphology and ultrastructure of blood vessel of brain and brain tissue in cerebrum. Cerebellum and hippocampus of rats of alcoholic group were test by both microscope and electron microscope. [Discussion] In alcoholic group rats were fed with gradually increasing dosage of alcohol under the condition that they can tolerate.The feeding frequency must be ensured.Thus we can both make this animal test model more definite and avoid mortality rate of the white rats with increasing dosage of alcohol blindly as sell as save the experimental time.During the experiment the rats of alcoholic group present symptoms of cerebral apoplexy and there are lesion in their brain tissue that we can observe with both microscope and electron microscope in the end of experiment. So this type of alcoholic animal test model is feasible. Pathologic changes were found in blood vessel and tissue of brain in alcoholic group even in the early time of alcoholism with microscope. Arteriolae of frontal lobe in alcoholic rats present that vessel wall turns a little thicker, lumen turns a little narrow, some endothelial cells fall off, internal elastic membrane appears some plicae. Quite a few smooth muscle cells become hyperplasia and cytoplasm turns a little hyperchromatic. Clooagenous fibers of external elastic membrane became hyperplasia, too.
