Background ApoE is a plasma protein that plays an important role in the metabolism of cholesterol and other serum lipids.It is an important determinant of risk for the development of atherosclerosis.Although the polymorphisms of ApoE gene has been found to be consistently associated with atherosclerosis and coronary heart disease,the relationship between ApoE polymorphisma and cerebrovascular disease is controversial.Objective To study the relationship between the polymorphisms of apolipoprotein E(ApoE),Cerebral infarct and intracerebral hemorrhage in naxi populations from Yunnan Province,and to compare with each group's difference in ApoE alleles in serum lipid levels.Methods We used PCR-RFLP method to detect ApoE genotypes of 90 patients with cerebrovascular diseases including CI group(58 cases),ICH group(32 cases), and that of age-and sex-matched normal group(50cases,male 27 cases,female 23 cases) in naxi populations,contrasted with the results.And the concentration of apoE in patients with CVD and in control subjects was determined.Meanwhile we detected their plasma lipid levels and hemorheological characteristics.At last,we used gene sequencing to verify the ApoE gene polymorphism with 20 cases(CVD patients with 10 cases,10 cases of normal controls).Results(1) The frequency of genotype E3/4 was 39.7%and e4 was 23.3%in Naxi CI group,significantly higher than those in Naxi control group(P<0.05).The frequency of genotype E3/3 was 48.3%in Naxi CI group,which were not significantly lower than in the Naxi control group(P>0.05).(2)There was no difference in the genotype frequencies and alloele frequencies of ApoE between ICH and controls.(3)Different cerebral infarction patient carry differentε4 allele frequency.they were atherosclerotic thrombotic cerebral infarction(55.3%),lacunar infarction(25.0%),and there was significant difference between atherosclerotic thrombotic cerebral infarction group and lacunar infarction group(P<0.05).(4)CI patients had higher levers of TG,TC,and LDL-c than the normal controls(P<0.05), There were no differences between ICH and controls in plasma lipid levels(P>0.05). (5)ε2 carriers had a higher levers of TG,HDL-c than theε3 andε4 carriers,andε4 carriers had a higher levers of TC and LDL-c than theε2 andε3 carriers(P<0.05 ).E2/4 carriers had a higher levers of TG thanε4 carriers(P<0.05).Conclusions(1) Our data show an association between ApoE gene polymorphism and the onset of CI and support the possibility that epsilon4 allele is a susceptibility locus for the risk of CI.But no evidence of relationship was found in apolipoproteinE genetype and cerebral hemorrhage.(2)The occurrence of stroke may be affected by interaction between serum lipids and ApoE gene polymorphisms.(3)ε4 allele is correlated with atherosclerosis related thrombosis,and irrelevant with lacunar infarction.(4)We used both PCR-RFLP methods and gene sequencing method to detect ApoE genotype,the results are simillar;but gene sequencing is more accurate,rapid,reliable,and missed the low rate of repeat good and so on.
