Cancer is one of the main diseases which do harm to the human healthy. It is urgent for us to control and cure it. How we treat cancer mainly depend on the combination use of operation, chemotherapy, radiotherapy and Chinese medicine. But we havn't effective method to detect earlier period cancer. Tumor markers are commonly used method to detect tumors in clinical. MUC1 is one of the tumor markers. MUC1 protein is not only highly expressed in tumor constitution but can be detected in blood serum in tumor patients. As so we regard MUC1 as a tumor marker and use it to diagnose and treat tumor. The immunohistochemical method is not fit for the patients who have no visible tumor after operation. If we can detect MUC1 in blood serum, it will be helpful for us to judge the effect of treatment and prognosi. In this study we use immunohistochemical method and di-antibody ELISA method to detect the changes of MUC1 protein in tumor tissue and in blood serum before and after chemotherapy and discuss the relations between these changes and the tumor therapeutic efficacy. Through this study we try to find reasonable methods and measures to diagnose tumor earlier or to judge the therapeutic efficacy and prognosi.1.By immunohistochemical method we detect the expression of MUC1 protein in breast and stomach benigh and malignant tissues. We find that the expression rate of MUC1 grow higher in breast benigh lesion, carcinoma and para-malignant tissues. The bigger the tumor is and with the more lymph nodes metastasis, the higher will MUC1 protein express in the tumor tissues. All these hint that MUC1 protein is related to the tumor metastasis and infiltration. The results that the MUC1 protein highly expressed in stomach carcinoma contrast to the paramalignancy hint that the expression of MUC1 in the stomach carcinoma tissues have nothing to do with the size of tumor, lymph node metastasis and clinical stage. We think that there have relations between the MUC1 expression in carcinoma tissues and blood serum MUC1 expression. So we can detect blood serum MUC1 to judge the tumor malignant degree and prognosi. 2.We detect the blood serum MUC1 of breast, stomach and ovarion benign and malignant patients with di-antibody IRMA method. The results show that there are no obvious changes between the benign lesion patients before and after operation the same as the tumor patients. The MUC1 in blood serum has significant difference between benign and malignant tumor patients. The MUC1 in blood serum obviously declined after chemotherapy in malignant tumor patients. The MUC1 in blood serum grow again in some patients with tumor recurrent and metastasis. All results show that the MUC1 in blood serum related to the progress of the tumor.This study show that MUC1 protein as a new tumor marker express highly in the blood serum of malignant patients especially in adenocarcinoma patients. We can use it to diagnose, treat and prognose the tumor. If the blood serum MUC1 of tumor patients grow continuously during or after chemotherapy we should be alert of the recurrent or metastasis of the carcinoma.
