ObjectiveThe epidemiological study suggests that the morbidity of coronary heart disease has the gender differences. Many experiments proved that the protective effects of estrogens accounted for it. But recent clinic study found that the hormone replace treatment doesn' t reduce the morbidity of coronary heart disease in menopausal women. This suggests that the androgen perhaps has some protective effects. So the human monocytic cell line THP-1 Cells were stimulated by testosterone in different concentrations, and the expression of Androgen Receptor and Intercellular Adhesion Molecule-1 were examined to explore the relationship of androgen and coronary heart disease.MethodsThe cultured THP-1 was stimulated by testosterone in different concentrations at different time. Extracting the protein and RNA from the THP-1, detecting the Androgen Receptor and Intercellular Adhesion Molecule-1's protein and RNA. The protein expression of Androgen Receptor and Intercellular Adhesion Molecule-1 was examined with Western Blot. The genetic expression of Androgen Receptor was examined with RT-PCR.Results1 There are AR in human mononuclear/ phagocyte. The quantity of AR's expression can be reduced by testosterone.2 Comparing with the control, the AR's expression can be reduced by testosterone at the concentration of KTmol/L to KT'mol/L, and there is concentration dependence from 10~9 mol/L to 10"7mol/L. The selective androgen receptor blocker flutamide can abolish this action. The AR protein was reduced more for stimulated 24hours than 12hours and there is time dependence too.3 Adding TNF- a can induce the ICAM-1 's expression increasing much more than the control, and this increasing can be inhibited by testosterone. After stimulating 12hours, the ICAM-l's protein expression increasing induced by TNF- a was reduced at the concentration of 10"7 mol/L to 10~5mol/L. After stimulating 24hours, the ICAM-l's protein expression increasing was reduced at the concentration of 10"8 mol/L to 10~5mol/L.Conclusion1 -. There are the AR in human mononuclear/ phagocyte. 2 The quantity of AR's expression can be reduced by testosterone. 3 TNF- a can induce the ICAM-1 's expression increasing. 4Testosterone can reduce the ICAM-l's protein expression increasinginduced by TNF- a .5 The selective androgen receptor blocker flutamide can abolish the testosterone's action.
