ObjectiveThe model of insulin resistance was made by high-fat diet.The level of insulin was detected and the expression of intercellular adhesion molecule-1,vascellum cellular adhesion molecule-1 in aterial endothelialitric and IL-18 in myocardial cell were observed.To observe the change of and intercellular adhesion molecule-1, vascellum cellular adhesion molecule-1,IL-18 which caused the changing in high-fat diet rats,and to evaluate its action in diabetic cardiomyopathy accelerated by insulin resistance.methods20 male Wistar rats were randomly assigned into 2 groups:normal diet group and high-fat diet group after 1-week-accommodation raising.Every rat was weight once a week,Each froup was fed for 8 weeks.IR was verified by euglycemic-hyperinsulinemia clamp technique,Myocardial tissue were taken for immunohistovhemistry detecion,Blood from left ventricle was taken for the detection blood serum's factors. Level of insulin was detemined and the expression of intercellular adhesion molecule-1, vascellum cellular adhesion molecule-1 in aterial endothelialitric and IL-18 in myocardial cell was examined by immunhistochemistry.The differences of these factors between the two groups and the coorelation between the factors linked to IR and atherosclerosis were respectively analyzed by statistics softwate SPSS13.0Result1.IR was verified in high-fat diet group by using euglycemic-hyper-insulinemia clamp technique,but not in normal diet group.In normal diet group,FBG was 5.01± 0.20mmol/l,the level of insulin was 1.41±0.44mmol/l and GIR was 14.41±0.69mg/kg.min.In high-fat diet group,FBG was5.33±0.20mmol/l,the level of insulin was 2.23±0.85mmol/l and GIR was 9.40±1.29mg/kg.min2.In high-fat diet group,the intercellular adhesion molecule-1,vascellum cellular adhesion molecule-1 in aterial endothelialitric and IL-18 in myocardial cells were significantly expressed in myocardial cells(P<0.05).But in normal diet group,there is little expression.3.Immunohistochemistry index:the expression of intercellular adhesion molecule-1 in aterial endothelialitric was positive correlated with expression of IL-18 in myocardial cells(p<0.05).and the expression of vascellum cellular adhesion molecule-1 in aterial endothelialitric wad positive correlated with expression of IL-18,too(p<0.05)Conclusions1.In insulin resitance,in high-fat diet group,the intercellular adhesion molecule-1 were significantly expressed in aterial endothelialitric,but in normal diet group,there is little expression.2 In insulin resitance,in high-fat diet group,the vascellum cellular adhesion molecule-1 were significantly expressed in myocardial cells,but in normal diet group, there is little expression.3.In insulin resitance,In high-fat diet group,the I1-18 were significantly expressed in aterial endothelialitric,but in normal diet group,there is little expression.4.Immunohistochemistry index:the expression of intercellular adhesion molecule-1 in aterial endothelialitric was positive correlated with expression of IL-18 in myocardial cells(p<0.05)).and the expression of vascellular adhesion molecule-1 in aterial endothelialitric wad positive correlated with expression of IL-18,too.(p<0.05))...
