Background and ObjectiveHuobahuagen tablet, a traditional Chinese medicine which is peeled skin and then whose root is made into medicine, belongs to celastraceae common threewingnut root genus. This traditional Chinese medicine grows in Xichang, Dechang, and liangshan etc, Sichuan province. It is a pure traditional Chinese medicine manufactured by Chongqi Traditional Chinese Medicine Research Institute. Huobahuagen tablet tastes bitter, acrid and chilly and it mainly comprises of alkaloid, lactone, phenic acid kind, pigment, and celastraceae alkaloid etc. Clinical studies suggested that Huobahuagen tablet had effective anti-inflammation, analgesia, and immune regulation, and had significant curative effect on several autoimmunity diseases such as psoriasis, chronic glomerulonephritis, rheumatoid arthritis and erythema nodosum. Huobahuagen tablet have no other side effects of glucocorticoid.Nowadays, researches suggest that asthma is a chronic inflammatory disease of the airway which is participated in mainly by eosinophil, mast cell, T lymphocyte, and other inflammatory cells. It is generally accepted that eosinophil maybe play the most important role in inducing inflammation of airway in all the inflammatory cells and its intensity of effect on asthma is the biggest in all the inflammatory cells. Activate T lymphocytes secrete interleukin3 (IL-3) and interleukin5 (IL-5) to promote eosinophil differentiation, activation, survival, and airway recruitment. Cytokine IL-3 and IL-5 have common B subunits which are the key point of signaltransduction, and they have many similar or same effect. These two cytokines play a part by combining together with respective a subunit of receptor of eosinophil.They promote eosinophil differentiation, ripe, adhesiveness of endothelium, activation, chemotaxis, and casting off granules. Activation, infiltration and releasing of eosinophil are the main causes to induce characteristic pathological changes and AHR. A report suggested that the increasing of expression of IL-3 and IL-5 of bronchoalveolar lavage fluid(BALF) was relevant to severity of asthma.We measured the total cell count and cell classification of BALF of guinea pigs, at the same time pulmonary histopathologic examination and Immunohistochemistry of IL-5 were performed. Finally, We investigated the expression of IL-3 receptor mRNA and IL-5 receptor mRNA in BALF after Huobahuagen tablet therapy and dexamethasone therapy. We made conclusions about the anti-inflammation mechanism of Huobahuagen tablet on the airway of asthmatic guinea pigs, and offered an experimental groundwork for application of this traditional Chinese medicine on clinical therapy of asthma.Materials and MethodsMaking of the model of asthmatic guinea pig and grouping.Thirty-two healthy, parti-colored guinea pigs were randomly divided into four groups in which each group had eight guinea pigs: control group, asthma group, dexamethasone therapy group and Huobahuagen tablet therapy group.Making of the model of asthmatic guinea pig: 1ml of 10% ovalbumin (OVA)saline suspension were injected into abdominal cavity of twenty-four guinea pigs of threes groups in the first day (The albumin flake was grand into fine powder before dissolution) . Then, aerosol of 1%OVA saline suspension was sprayed to challenge these sensitive guinea pigs by ultrasonic atomizing inhalation instrument in the fifteenth day. We stopped challenginguntil we found guinea pigs breathing quickly, nodding spasm, and rectus abdominis shrinking. From above three groups, one group was asthma group, the other two groups were treated by dexamethasone and Huobahuagen tablet respectively for two weeks. The last group was control group.Collection of BALF and measurement of the total cell number and cell classification.At the thirtieth day, we anesthetized them after the last challenge. Then, we poured 3ml Hanks into bronchus and lung, and retrieve BALF. We lavaged four times in all. BALF was centrifugated, and we took few of them to slide for smear and measured the total cell...
