THESIS 1. The experiments of separation and purification of pigpancreas islet[Abstract] Objective: To separate and purify the pancreas islet of adult pig then to measure its biological activities in order to provide adequate and high qualitative cells of pancreas islet to satisfy the need for transplantation Method: Islets were isolated using digestion by ductal injection of collagenase and purified by centrifugation on Ficoll gradients. The activities detection of pancreas islet cells(PIC) were measured by insulin release stimulated by glucose. Result: The cellular purification of pancreas islet was about 85% +4%. The output of pancreas islet cells was near (4-7) X 107cells/pig. Otherwise after purification the numbers of pancreas islet cells were 1367+1876/g(pancreas). The activities of pancreas islet cells were 95% +3%. By the end of one day culture ,the secretive volumes of insuline of PIC stimulated by low concentration solution of glucose (5.5mmol/L) and high concentration solution of glucose (16.7mmol/L) were 408.23 + 56.40mU/L and 890.20 + 116.43mU/L respectively. Nevertheless, by the end of five-day culture the secretive volumes were 396.94 + 32.27 mU/L and 852.66 + 111.45 mU/L respectively. Based on statistical analysis, there was a significant different between one-day high glucose concentration group and low glucose concentration group(p<0.01). There was a significant different between five-day high glucose concentration group and low glucose concentration group(p<0.01) too.However there were not significant differences between high glucose group in one-day culture and high glucose group in five-day culture as well as between low glucose group in one-day culture and low glucose group in five-day culture respectively. Conclusions: The data associated with separation and purification of PIC of pig were provided by this experiment. Its purificatory degree and biological activities were satisfied so that the pancreas islet of adult pig can be a donor to use for transplantation.
