Objective:To examine the change of Interferon-γandInterleukin-4 in bronchoalveolar fluid from rats with pulmonaryfibrosis on different times after bleomycin-treated and tocompare with the saline control group ,in order to explore theeffect of TH1/ TH2 imbalance in pulmonary fibrosis .To examinethe change of Interferon-γ, interleukin-4 and cell differentialanalysis in bronchoalveolar fluid from BCG-PSN-treated ratswith bleomycin-induced pulmonary fibrosis and to comparewith the bleomycin-treated group,in order to explore theintervention of BCG-PSN on TH1/ TH2 imbalance . Methods: 43 male SD rats were randomly divided intotwo experimental groups:the saline control group and thebleomycin A5(BLM) model group ,which was performed by thesingle transtracheal injection of bleomycinA5 and randomlydivided into model Ⅰ group(BLM group) and model Ⅱgroup(BLM+BCG-PSN group). ON 7,14,28 day afterbleomycin instillation,lung pathologies were gained andbronchoalveolar fluid were performed. Results:1.Concentrations of IFN-γ protein in BALF fromrats(pg/ml):in B1 and B2 group (BLM group) , the levels ofIFN- γ were 58.167 ± 9.227,86.022 ± 10.997,which weresignificantly lower than that in saline control group (131.033±19.261) (P ﹤ 0.001) ;There were no statistic significantdifferences between B3 group and A group. In C1(BLM+BCG-PSN) group, levels of IFN- γ (104.047 ±17.012)increased compared with B1 group (P﹤0.001), butdecreased compared with A group(P﹤0.05). In C2 group, levelsof IFN-γ(171.667±29.930) were significantly higher than thatin B3 group(P﹤0.001), and higher than that in saline contralgroup(P﹤0.01).2.Concentrations of IL-4 protein in BALF fromrats(pg/ml):levels of IL-4 in B1 group and C1 group were 11.15± 1.415 , 10.38 ± 1.037, which were significantly highercompared with A group(7.155±1.001)(P﹤0.001), but therewere no statistic significant differences between B1 group andC1 group. levels of IL-4 in B2,B3,C2 group were 7.71±1.052,7.225±1.051,6.80±0.635,which were no statistic significantdifferences compared with A group,and the same results wereseen between C2 group and B3 group. 3.Total cell counts anddifferent analys in BALF from rats: The total number ofcells(109/l) in B1,B2,C1 group were 7.8±1.37,4.7±0.62,7.2±1.43, which were significantly higher than that in saline 5英 文 摘 要control (1.2±0.32) (P﹤0.001). In B1,B2,C1 group,theconcentrations(%)of neutrophils and lymphocytes significantlyincreased than that in A group , in contrast , theconcentrations(%) of macrophages decreased than that in Agroup((P﹤0.001)。B3,C2 group compared with A group,C1group compared with B1 group,C2 group compared with B3group,there were no statistic significant differences in total cellcounts and differential analysis. 4.Semi-quantitative analysis ofthe histopathologic changes: Lower degrees of lesions were seenin BLM+BCG-PSN groups compared with their BLM controls,but there were no statistic significant diffierences. Conclusion: 1.There is "TH2" predominant in BALF frombleomycin-induced pulmonary fibrosis in rats, namely,theconcentration of IFN-γ is decrease and the concentration ofIL-4 is increase.2.BCG-PSN could increase the inducation ofIFN- γ and incompletely shift the TH1/ TH2 balance.3.BCG-PSN propably have an assistant effect on the fibroticprocess induced by BLM in rats.
