Objective: This experiment establish rat's slow transit constipation(STC) model using rhubarb ,coping pathological process after taking medicine and observing its colon transmit function, pathological change of colon wall, VIP and SP's immunoreactive recovery in colonic myenteric plexus. By means of research in Fushu capsule, we observe colon transit function, pathological change, ultrastructure change of colon wall, vasoactive intestinal peptide (VIP) and substance p(SP)'s change in colonic myenteric plexus and relativity of VIP and SP's immunorective change in colonic myenteric plexus and pathological change in colonic myenteric plexus. In addition we discuss mechanism of Chinese traditional medicine preventing and treating STC.Material and method1 Animal model preparation1.1 Animal groups distribution: choose 72 clean healthy adult SD rats, female and male half respectively, these rats were divided randomly into control group and rhubarb group, no marked weight difference after grouping. 1.2 Method: Feeding control group with ordinary feedstuff; Feeding rhubarb group with feedstuff containing rhubarb. For the beginning the dosage is 100mg/kg.d, afterwards the dosage double every day until half of rats' stool become watery, maintaining this dosage until 80 percent of rats' watery stool disappear, in this base doubling dosage until half of stool become watery. Thus this procedure circles three times, then stopping feeding one week after the last 80 percent of watery stool disappear waiting for treatment feeding with ordinary feedstuff.2 Animal groups distribution and treatment: Rhubarb model rats were divided randomly into 5 groups, random one group of rhubarb model rats was chosen to measure intestinal transmit rate compared with normal group; Thus observing pathological and ultrastructural change in colon wall by means of HE staining, argentophil staining, immunohistochemical staining and electronic microscope. The residual 4 rhubarb groups were respectively administrated fushu capsule , tongbianling capsule, physiological saline by stomach perfusion for 2 weeks. Measuring intestinal transmit rate using active carbon liquid pressing; Thus observing pathological and ultrastructural change in colon wall by means of HE stain, argentophil stain, immunohistochemical stain and electronic microscope.3 Data processing: This experiment's measure data is expressed by xˉ±s, adopting SPSS 10.0 stat software, choosing two-group one-way ANOVA, test level α=0.05;Result of colon transmit function, surface density result of VIP,SP positive nerve immunohistochemistry in colonic myenteric plexus, offer total data, adopt SPSS10.0 stat software, choose two-group one-way ANOVA, test level α=0.05;Half quantitative result of VIP,SP positive nerve immunohistochemistry in colonic myenteric plexus, test method is two-group multiple-samples rank sum test(Kruskal-Wallis Test)and multiple comparision(Nemenyi Test),test level α=0.05.Result1 In this experiment animal weight in rhubarb group is lower than that in control group, animal weight in treatment group varies little, there is marked difference compared with normal group(p<0.05). The appearance of intestines vary little among groups and there is no pigmentation in colonic mucosa .2 Intestinal transmit function: Compared with control group transmit function evidently become slower in rhubarb group(p<0.01); Intestinal transmit of single fushu capsule group, triple fushu capsule group, tongbianling capsule group and natural recovery group is evidently quicker than that of rhubarb group(P<0.01);Intestinal transmit of single single fushu capsule group,triple fushu capsule group,tongbianling capsule group is evidently quicker than that of natural recovery group(P<0.05);Intestinal transmit is no marked difference among single fushu capsule group, triple fushu capsule group, tongbianling capsule group. 3 Result of optical and electronic microscope3.1 Optical microscope(HE staining): In the rhubarb group chronic inflammation in mu...
