Objective: The orthotopic transplantion tumor model and the subcutaneous ectopia transplantion tumor model of hepatorcellular carcinomas were established in mice by injecting the H-22 hepatocarcinoma cell, to explore the factor of affecting SPIO-enhanced MRI signal change of hepatocarcinoma in clinic .Materials and methods: (1) The clinical -. imageing and pathological data of SPIO-enhanced MRI signal change of 25 cases with hepatopathy was analysed; (2) Fourty orthotopic transplantion tumor models and fourty subcutaneous ectopia models of hepatorcellular carcinomas in mice were maken by injecting the H-22 hepatocarcinoma cell; (3) The transplantion mices were scaned before and after SPIO-enhanced MRI; (4) HE and prussian blue staining in all transplantation mice was done and analysed the result of the staining.Results : (1) Of 25 cases , there were 2 hepatocarcinoma cases 1 angiocavernoma case and 1 cirrhosis of liver node case diagnosed by pathology, and 2 cases have typical sign by DSA and interventional therapy; (2) The T2WI signal of pre-enhanced and post-enhanced by SPIO had not changed significantly in the 40 case of the subcutaneous ectopia transplantion tumor models; prussian blue staining was positive and the macrophage can be seen in the inner of tumor lesion in two transplantion mices ;(3) T2WI signal intensity of post-enhanced by SPIO of 10 of 40 orthotopic transplantion tumor models had not change significantly and tumor lesions is clear to the nomal tissue of liver; pathological prussian blue staining was positive and the kupffer cell can not be seen in tumor lesion except for 3 transplantion mices , infiltration growth between tumor lesions and nomal liver tissue can be seen in 7 transplantion lesions; degeneration and necrosis tissue can be seen in 13 transplantion lesions.Conclusion: The diagnosis rate of SPIO to occupying lesion of liver was high, but the change of SPIO-enhanced MRI signal did not match with pathological mechanism; transplantion tumor models in mice by injecting the H-22 hepatocarcinoma cell could be applied to the experiment study of SPIO-enhancedMRI signal change ; there were some kupffer cells in the inner and edge of several transplantion tumor lesions, and it was the important cause to change the enhanced transplantion tumor MRI signal ; it was related factor to change the SPIO-enhanced transplantion tumor signal to part of transplantion tumor growth manner and the nomal liver tissues and degeneration and necrosis in the inner of transplantion tumor.
