PART1The Study of the Targeting of c-erbB2AntisenseProbe Labeled with Superparamagnetic Iron OxideNanoparticles Used for MR of BALB/c MiceObjectiveTo investigate the targeting of c-erbB2antisense probe labeled withsuperparamagnetic iron oxide nanoparticles used for Magnetic resonanceimaging of BALB/c mice.MethodsGet ready the establishment of SK-Br-3tumor-bearing BALB/c mice10, each mouse injected with antisense probe (12.0mg Fe/Kg); Each mousehad MR scan in the given point of time (before injection and60、180、360、720min after injection); After MR scan, take the liver、spleen、kidney andtumor tissue of the mice immediately, make the tissue fixed by10%formalinand paraffin-embedded sections, HE and Prussian blue staining, observed under microscope; Measure the signal strength of liver、spleen、kidney、tumor tissue and adjacent muscle, compared the signal intensity changes inthis tissue.ResultThe rate of establishment of SK-Br-3tumor-bearing BALB/c mice is100%. Contrast the signal change curve of liver、spleen、kidney、tumortissue and adjacent muscle after injection of antisense probe, found that thetumor signal change obviously lagged behind the rest, there has obviouslydifferences exist in signal change curves.ConclusionThe c-erbB2antisense probe labeled with superparamagnetic iron oxidenanoparticles can be targeted action on tumor tissue of BALB/c mice.PART2The Investigation on the Optimal Scan Time ofc-erbB2Antisense Probe Labeled with SuperparamagneticIron Oxide Nanoparticles Used for MR of BALB/c MiceObjectiveTo investigate the optimal scan time of c-erbB2antisense probe labeledwith superparamagnetic iron oxide nanoparticles used for Magneticresonance imaging of BALB/c mice. MethodsGet ready the establishment of SK-Br-3tumor-bearing BALB/c mice20; Each tumor-bearing BALB/c mouse had MR scan before injection; Eachtumor-bearing BALB/c mouse Injected with antisense probe (theconcentration is12.0mg Fe/Kg), had the MR scan in different time points(60min,180min,360min,720min) after injection and immediately take thetumor tissue of4mice, make the tissue fixed by10%formalin andparaffin-embedded sections, HE and Prussian blue staining, observed undermicroscope; Measure the signal strength of tumor and adjacent muscles,compared the signal intensity changes in different time points.ResultThe rate of establishment of SK-Br-3tumor-bearing BALB/c mice is100%; Contrast the signal intensity of20mice at different time points beforeand after injection, the signal intensity changes of the tumor issue in360minafter injection are most obvious. The pathological examination revealed thattumor issue structural disorder, there are a large number of special-shapedhumor cell exist, arranged in the cancer nest shape, and a large number ofpunctuate blue iron particles scattered in the tumor issue in each time point.And the distribution of iron particles is most densely in360min afterinjection.ConclusionThe optimal scan time of c-erbB2antisense probe labeled withsuperparamagnetic iron oxide nanoparticles used for Magnetic resonance imaging of BALB/c mice is360min.PART3The investigation on the optimal concentration ofc-erbB2antisense probe labeled with superparamagnetic ironoxide nanoparticles used for MR of BALB/c miceObjectiveTo investigate the optimal concentration of c-erbB2antisense probelabeled with superparamagnetic iron oxide nanoparticles used for Magneticresonance imaging of BALB/c mice.MethodsGet ready the establishment of SK-Br-3tumor-bearing BALB/c mice20, randomly divided into5groups and each group injected with differentconcentrations of antisense probe (6.0mg、9.0mg、12.0mg、15.0mgFe/Kg); Each group had MR scan in the given point of time (before injectionand360min after injection); After MR scan, take the tumor tissue of the miceimmediately, make the tissue fixed by10%formalin and paraffin-embeddedsections, HE and Prussian blue staining, observed under microscope;Measure the signal strength of tumor, compared the signal intensity changesin different groups.ResultThe rate of establishment of SK-Br-3tumor-bearing BALB/c mice is 100%; The BALB/c mice of6.0mg、9.0mg and12.0mg group were all aliveafter MR scan, but part of the BALB/c mice of15.0mg group were deadbecause of the excessive Concentration. Contrast the signal intensity of eachgroups at different concentrations before and after injection, the signalintensity changes of the tumor issue in12.0mg、15.0mg group are obvious,and the signal changes of the tumor tissue has been visually identification;there was only a little difference among the three groups. The pathologicalexamination revealed that tumor issue structural disorder, there are a largenumber of special-shaped humor cell exist, arranged in the cancer nest shape,and a large number of punctuate blue iron particles scattered in the tumorissue in each concentration. And the distribution of iron particles is denselyin12.0mg、15.0mg group after injection.ConclusionThe optimal concentration of c-erbB2antisense probe labeled withsuperparamagnetic iron oxide nanoparticles used for Magnetic resonanceimaging of BALB/c mice is12.0mg Fe/Kg.
