Objective: to study the infiltration of DCs and expression of IL-10 and their roles in antitumor immunity in primary gallbladder cancer, intended to properly understand the poor prognosis of such malignancy in immune perspective.Methods: 56 specimens, first of all, any diseases combined affecting human being immune function was excluded, histological diagnoses disclosed various lesions of gallbladder, 37 of them diagnosed as primary gallbladder cancer(11 male cases, 26 female cases and aged under 60-year 9 cases, over 60-year 28 cases), other 19 cases including chronic cholecystitis(10 cases) and adenoma of gallbldder(9 cases), were performed using monoclonal antibodies against cell markers, S-100, CD1a, and IL-10 by immunohistochemistry. Quantitively analyze the infiltration of DCs and expression of IL-10 in various lesions of Gallbladder epithelial, SPSS program was employed to test statistical significance of the results. Results: The case percentages of S-100 and CD1a positive DCs in the tissues of Gallbladder cancer were lower than those in other benign lesions of Gallbladder(P<0.05), and the number varied with tumor differentiation type, invasion depth and lymph node metastases of the tumor(P<0.05); Expression of IL-10 in tumor tissues was higher than that in benign diseases(P<0.05), but not varied with tumor differentiation, invasion depth or lymph node metastases(with a nearly common concentration)(P>0.05); As to the relationship between DC infiltration and IL-10 expression in tumor tissues, no certain correlation was found(P>0.05).In addition, DC infiltration and IL-10 expression in tumor tissues have no relation with the gender and age of patients(P>0.05). Conclusions: DCs play positive important role in tumor immunity, their function and number in primary gallbladder cancer are key in initiating tumor specific immunity and may be used to estimate its prognosis; While IL-10 expression levels increased in tumor tissues, it plays negative role in tumor immunity commonly through forming a inhibiting microenvironment. The immune capability of the patients with malignancy did not vary with different gender or age.
