The Clinic Curative Effect Observation Of Jian Pi Yi Qi Prescription Treating To CAG & PLGC And Research Of Antitumor Immunity

Objective To observe clinical curative effect on precancerous lesions of gastric cancer treated by JianPiYiQi prescription, and through experiments in vitro, explore the role of Chinese medicine monomer—Astragaloside IV with DC vaccine in cell immunologic function and antitumor effect.Methods Clinical part:60 cases of the patients of precancerous lesions of gastric cancer were randomly allocated to treatment grope (30 cases) treated by JianPiYiQi prescription, and control group (30 cases) treated by WeiFuChun, and observe the clinic effect after 3 months treatment. Experiment part:Peripheral blood mononuclear cells (PBMCs) from healthy volunteers were isolated, cultured and generated in vitro, pulsed with tumor antigen from SGC7901 gastric carcinoma cell lysates, produced DC vaccine. Observe T-cell proliferation responses stimulated by DC vaccine with AⅣgroup, DC vaccine group and AⅣgroup respectively, and the anti-tumor effects on SGC7901 cells in vitro.Results Clinical part:the results showed that total effective rate in the treatment group and the control group was 86.7%and 63.3% respectively, the difference had statistical significance(P<0.05).The treatment group had symptomatic improvement in abdominal mass,gastric abscess,anorexia,hypodynamia and diarrhea significantly better than control group(P<0.05or P<0.01). The treatment group patho-atrophy,intestinal metaplasia improvement rate was 83.3%,80.8%, while the control group was 56.7%,51.9% respectively, the difference had statistical significance (P<0.05). The two groups atypical hyperplasia improvement rate was 60%,50%respectively,the difference had no statistical significance(P>0.05).Experiment part:①Lymphocyte proliferation:The T-cell proliferation rate of DC vaccine with AⅣgroup and DC vaccine group were significantly higher than AⅣgroup and T-cell group (negative control group) (P<0.01).The stimulating efficacy on T-cell proliferation of DC vaccine with AⅣgroup was higher than that of DC vaccine group (P<0.05).Compared with T-cell group, the T-cell proliferation rate of AⅣgroup had no statistically significance (P>0.05).②Killing effect on target cells:the killing rate of effector cells actived by DC vaccine with AⅣgroup and DC vaccine group against SGC7901 gastric carcinoma cells were higher than that of AⅣgroup and T-cell group (P<0.01).The killing power of DC vaccine with AⅣgroup was stronger than that of DC vaccine (P<0.05); while the group of AIV group and T-cell group cannot killing tumor cells. Both had no statistically significance (P>0.05).Conclusion 1. JianPiYiQi prescription treating to CAG & PLGC possesses a good curative effect.lt can improve the clinical symptoms and pathological conditions.2. AⅣcan stimulate T-cell proliferation and enhance the activity of killing tumor cells by DC, which induce specificantitumor response against stomach carcinoma cells effectively.
