Objective: To observe the expression of COX-2,c-erbB-2 and Ki67 in carcinoma, adenoma and normal mucosa in large intestine. To evaluate the relationship between the expression of COX-2,c-erbB-2, Ki67 in colorectal carcinoma and the clinicopathological parameters and to analyze the correlativity of the genes. To investigate the effect of COX-2,c-erbB-2, Ki67 in the occurrence and development of colorectal cancer in order to provide some theoretical basis for the clinical diagnosis and genetic therapy of colorectal cancer.Methods: The S-P immunohistochemical method was used to detect the expression of COX-2,c-erbB-2 and Ki67 in 11 normal colorectal mucosa, 11 colorectal adenoma and 51 carcinoma. Results were analyzed by SPSS version 10.0.Results: (1)With the ailments developing seriously, the level of positive expression of COX-2,c-erbB-2 and Ki67 in normal colorectal mucosa, colorectal adenoma and carcinoma increased gradually, significant differences were observed (P<0.001; P<0.05; P<0.01). (2) The positive expression of COX-2, c-erbB-2 in colorectal carcinoma was related with lymph nodes metastasis and Duckes stage: higher in stage C and D than stage A and B (P<0.05); lower in non lymph nodes metastasis group than metastasis group (p<0.05). Both had no relationship with histological grade. Ki67 wasn't related with histological grade, lymph nodes metastasis and Duckes stage. (3) In colorectal carcinoma the expression of COX-2 was positively related with that of c-erbB-2 and Ki67 (p<0.01), the expression of c-erbB-2 was positively related with that of Ki67 (p<0.05). (4)To analyze COX-2,c-erbB-2 and Ki67 together , results showed that the Ki67 average labeling index of group COX-2(+)c-erbB-2(+)was significantly higher than that of group COX-2(+)c-erbB-2(-), group COX-2(-)c-erbB-2(+), group COX-2(-)c-erbB-2(-)(p<0.001); between group COX-2(+)c-erbB-2(-) and group COX-2(-)c-erbB-2(+), there was no significance (p>0.05).Conclusions: (1)The positive expression of COX-2,c-erbB-2 and Ki67 may have an important role in the pathogenesis and development of colorectal carcinoma. (2)The combination of the expression of COX-2,c-erbB-2 and Ki67 may be helpful for judging the cell proliferation, malignant degree and prognosis. (3) Selective COX-2 and c-erbB-2 inhibitors may do a lot for the prevention and therapy of colorectal carcinoma.
