AIM: To investigate the protective effect of ischemic preconditioning against gut barrier function injury after rat small intestine ischemia/reperfusion and serious cutis burn .Methods: Wistar rats (♂, n=60, 240-280g, provided by Animal Science Partment of Jiangxi Medical University ) were fasted for 24 h before the experiments with access to water. The rats were randomly divided into 5 groups, there are 6×2 rats in each group. ⑴Control (C) group:only laparotomy was performed without clipping of SMA; ⑵Ischemic /reperfusion (IR) group:rats were subjected to 30-min occlusion of SMA followed by 60-min reperfusion; ⑶Ischemic preconditioning +Ischemia /reperfusion (IP+IR) group; the same as IR group,but with previous 3-cycle occlusion of SMA for 8-min and 10-min reperfusion; ⑷ Burn (B) group: rats cutis of 30% TBS area were burned by Electrical scald instrument with constant temperature and pressure of 106℃,0.03Mpa for 9s.The sample were detected for deep third degree by pathological examination. ⑸ Ischemic preconditioning +Burn(IP+B) group: the same as B group, but with previous 3-cycle occlusion of SMA for 8-min and 10-min reperfusion. After the specimen obstained, HE and PAS and AKP and IHC staining techique was used. PAS staining technique was used to study the integrality of intestinal epidermal basement membrane and the thickness of intestinal villi sugarcoating. On the other hand, HE staining technique and Chiu CJ grade were used to appreciate the pathological changes of damaged intestine in each group. The diamine oxidase (DAO) activity of intestinal tissue and blood was mesured with spectrophotometry in our experiment. The stereological parameter varities of Microvessel and the density varities of CGRP-ir,NPY-ir positive peptide nerve fiber in intestinal villi were measured by stereological method. Results: It was shown that: ⑴The thick red sugarcoating were seen to cover on the surface of intestinal villi and the basement membrane were intergral and successive in control group. In IR and B and IP+B group, the whole sugarcoating on intestinal villi surface were falled off and the basement membrane were broke to varying degrees, which was the most serious in IR group.In IP+IR group,the sugarcoating on intestinal villi surface was not dropped but only thinner,and its basement membrane was intergral too. ⑵Compared with control group, Chiu CJ grade of intestine in each damaged group were all impoved (P <0.01), which was the highest in IR group. Compared with IR group, Chiu CJ grade of intestine in IP+IR group were singnificantly decreased (P <0.01). ⑶Compared with C group, the DAO activity of blood in IR or IP+IR or B or IP+B groups was significantly improved but the DAO activity of small intestinal mucous decreased (P <0.01). Compared with IR group, the DAO activity of blood in IP+IR group was significantly decreased but the DAO activity of small intestinal mucous was significantly improved (P <0.01). ⑷Compared with C group, the mean section area(Aˉ ) and mean circumference(Bˉ )and length density(Lv) and volume density(Vv) of Microvessel in IR or IP+IR or B or IP+B groups were significantly decreased respectively (P <0.01and P <0.05). Compared with IR group, the mean section area(Aˉ ) and mean circumference(Bˉ )and length density(Lv) and volume density(Vv) of Microvessel in IP+IR group were significantly improved respectively (P <0.01). ⑸Compared with C group, the density of CGRP-ir positive peptide nerve fiber in intestinal villi was significantly decreased and NPY-ir positive peptide in intestinal villi nerve fiber was significantly improved in IR or IP+IR or B or IP+B groups (P <0.01). Compared with IR group, the density of CGRP-ir positive peptide nerve fiber in intestinal villi (P<0.01),was significantly improved but NPY-ir positive peptide nerve fiber in intestinal villi was significantly decreased in IP+IR group (P <0.01). ⑹Compared with B group, the difference of IP+B group in all the above-mentioned contents have no statistic significance(P >0.05).
