Part One Establishment Of Subcutaneous Xenotransplanted Model OfHuman Epithelium Ovarian Cancer In Nude MiceAnd Its Biological CharacteristicsObjectiveTo establish a xenotransplanted subcutaneous tumor model of human epithelium ovarian carcinoma in nude mice for the in vivo research of human ovarian cancer in diagnosis and therapy.MethodsCulture and collect the SKOV3 cells in vitro and adjust the cells to available concentration. Inject the cells suspension subcutaneously into the nude mice. Then observe the growing of tumor. When it reached the size of 0.8cm~1cm in diameter,cut it off and into pieces of 1mm~2 mm in diameter,then it was transplanted subcutaneously into another nude mice.Observe all the tumor in pathology and electron microscopy. Chromosome and tumor marker ,such as CA125, AFP and CEA,were also analyzed. ResultsA subcutaneous xenotransplanted model of ovarian cancer was established successfully in nude mice ,and it had been passaged in vivo for two generations.Histological and electron microscopic observation, chromosomal karyotype were similar to human epithelium ovarian cancer.But metastasis of tumor was not observed in nude mice.The concentration of tumor marker CA125 is low in the serum,however it is high in the supernate of the rupture product of tumor. Conclusion The subcutaneous xenotransplanted SKOV3 model in nude mice has the characteristics of human epithelium ovarian cancer. It can provide a useful tool for the study of biology,diagnosis and therapy of human ovarian carcinoma.
