Purpose: 1.To design and produce a kind of mechanical property device for artificial dermais and then to measure the artificial dermaical mechanical property; 2.To detect the vitodynamics property of composite chitosan artificial skin after freezed and preserved; 3.To transplant the composite chitosan artificial skin over the ulcer of the volunteer and observe clinical changes.Methods: 1. According to the principle of fluid static pressure methods, along with loading the indirect pressure method. 2. Measuring the tensile strength of composite chitosan artificial skin preserved in various condition of temperatures with the equipment. 3. With the method of tissue engineering, producing the stand-in of Composite chitosan artificial dermis, and obtaining composite chitosan artificial skin. After skin organotypic culture, transplanting it to the ulcer of the volunteer, observing clinical changes of tissue engineered skin.Results: 1. To prepare the detecting equipment which have satisfied organism material mechanics property of high acuity micro- tension lemma. With the equipment, it can detect artificial skin mechanics property.2. Composite chitosan artificial skin have better tensile strength, and their mechanics property (standing pressure) are stable.3. After resuscitating, composite chitosan artificial skin frozen and preserved on various temperature conditions have the similar property as those have not frozen.4. Covering with the composite chitosan artificial skin on wound in patient, no case showed red swelling of the skin around wound. It was also found that no case emerged with empyema, hemorrhage and infection, no rejection reaction happened obviously. Composite chitosan artificial skin have better tensile strength, their mechanics property are stable. Composite chitosan artificial skin which have been frozen and preserved on various temperature conditions suit to meet an emergency in clinical demand. The composite chitosan artificial skin we establish reconstruct epidermis and dermis with favorable physics characteristics and can cover the full-thickness excised wound effectively.
