Objective: To study the basic mechanism and effect of Chitosan Chinese-medicine multiple membrane treating chronic skin ulcer on the molecular level. Methods: Back ulcer models were established on rats with literature. Then model rats were divided into three groups and treated by Chitosan Chinese-medicine multiple membrane, Physiology and shengjiyuhonggao respectively. Adopted the tissues of ulcer surface separately and observed their situation of inflammatory exudate and necrosis, granulation tissue's vegetation, macrophage exudate quantity, fiberic hyperplasia and epithelial migration by light microscope ,observed the form of neonatal basal cell and capillary vessel by electric microscope, observed the expression quantity of Basic fibroblast growth factor, Transforming growth factor by Power Vision Two-Step Histostaining method. Results: Chitosan Chinese-medicine multiple membrane can reduce inflammatory exudate and necrosis, increase granulation tissue's vegetation, macrophage exudate quantity, fiberic hyperplasia and epithelial migration. It has increased the expression quantity of Basic fibroblast growth factor and Transforming growth factor obviously, made it equal or more than shengjiyuhonggao group in the same period and they were different compared between the two groups (P<0.05) , it was different obviously compared with the Physiology group (P<0.01) too. Conclusion: ① Chitosan Chinese-medicine multiple membrane's collocation is reasonable and its using is convenient, the effect of treatment is equal or beyond shengjiyuhonggao group in the same period. (2) It can increase the expression quantity of BFGF and TGF- β1, so as to accelerate aggutination of the ulcer.
