Objective and Significance: The migration and drug resistance of leukemia cell and the infiltration of lymphoma cell are the basic characters of these two malignant hematological tumors, and they are also the basic reasons for the bad outcomes of the diseases. The SDF-1/CXCR4 axis, which consists of stromal cell derived factor-1 and its only receptor CXCR4, may play an important role in the above mentioned characters of the malignant hematological tumors via its function of changing the microenviroment of bone marrow and lymphatic system. Furthermore, the performance of the SDF-1/CXCR4 axis' biological function may be correlated with the expression of vascular endothelial growth factor(VEGF), the main factor involved in angiogenesis of many malignant tumors. In this topic we detected the expression level of SDF-1,CXCR4,VEGF of the above two malignant hematological tumors as well as their relations to the tumor cell's characters of drug resistance and infiltration etc, and then elucidated the change of the microenviroment' on the occurrence and development of hematologic tumor. This topic may provide some theoretical and clinical proofs for finding out some new therapeusis to the above two malignant hematological tumors.Materials and Methods: 26 patients with newly diagnosed acute leukemia and 18 patients with newly diagnosed lymphoma were choosed for study objects, 18 healthy adults were choosed for controls. Their bone marrow and peripheral blood were collected for detection. The expression of SDF-1 and VEGF in serum were detected by ELISA, and the expression of CXCR4 on MMNC's membrane was detected by FACS. The patients' clinical materials about tumor classification, infiltration and remission degree were also collected. All the data were analysed by SPSS statistic tool.Results:1. The expression of SDF-1, VEGF and CXCR4 in acute leukemia group was markedly higher than the control group, the patients with extra-marrow infiltration or poor therapeutic efficiency had high expression level of SDF-1 and CXCR4, while the high expression of VEGF was only observed in the patients with extra-marrow infiltration. The expression of SDF-1 and CXCR4 in ALL group was markedly higher than the ANLL group(P<0.01), while the expression of VEGF in ALL group was markedly lower than the ANLL group(P<0.01).2. The expression of SDF-1, VEGF and CXCR4 in malignant lymphoma group was markedly higher than the control group, the expression level of SDF-1 in the clinical stage was markedly higher than the clinical stage(P<0.01),while this markedly difference didn't lie in the expression of CXCR4 and VEGF. The lymphoma patients with bone marrow infiltration had high expression of SDF-1 and CXCR4.3. The correlation analysis showed that there was a markedly correlation between the expression level of VEGF/CXCR4 and VEGF in ALL and ANLL group, while in the lymphoma group, the correlation was not definite.4. The ROC curve of SDF-1 and CXCR4 for diagnosing acute leukemia was up protruding, the proportion of below-curve was over 0.5.Conclusion:1. Acute leukemia patients have high expression level of SDF-1, CXCR4 and VEGF, the expression of SDF-1 and CXCR4 can be used as an important diagnosis index for acute leukemia.2. The expression level of SDF-1 and CXCR4 in acute leukemia is associated with the type of blast cell.3. The high expression of SDF-1/CXCR4 axis is associated with the angiogenesis of the malignant tumors, some feedback modulation mechanism may be implied in the SDF-1/CXCR4 axis and the expression of VEGF.4. The high expression of SDF-1 and CXCR4 may have some correlation with the infiltration and drug resistance of acute leukemia.5. SDF-1, CXCR4 and VEGF are the key factors to malignant lymphoma, the development of malignant lymphoma is associated with the angiogenesis, the high expression of SDF-1 and CXCR4 can be used as diagnostic factors for judging the infiltration and stagesof Malignant lymphoma.
