Objective:To investigate the changes of bone marrow-derived stem cells (BMSC)and stromal cell-derived factor-1α(SDF-1α)and vascular endothelial cell growth factor(VEGF)in peripheral blood of patients with acute myocardial infarction(AMI)and explore their role in AMI.Methods:All the experimental patients were divided into three groups,which include AMI group(30 patients with AMI),SA group(30 ones with stable angina)and Control group(30 healthy adults).To collect peripheral blood of the patients in SA and control groups once and in AMI group third times, respectively on the first,third and seventh days after AMI had happened,as the samples.Flow cytometry was used to measure CD34+BMSC in peripheral blood and Enzyme-linked immunoassay(ELISA)was employed for measuring VEGF and SDF-1αin blood plasma.Results:(1)The concentration of CD34+BMSC in every sample of AMI group were obviously higher than that of SA group(P<0.05)and control group (P<0.05).However,there was no statistic significance about the comparison between SA and control group(P>0.05).(2)The concentration of CD34+BMSC on the seventh day of AMI group were obviously higher than those on the first and third day of the same group(P<0.05).However,compared the concentration between on the first and on third day,there was no statistic significance (P>0.05).(3)The concentrations of VEGF and SDF-1αon the 1st,3rd and 7th of AMI group were obviously higher than SA group and control group(P<0.05). However,there was no statistic significance about the comparison between SA and control group(P>0.05).(4)The concentration of VEGF on the seventh day of AMI group were obviously higher than those on the first and third days of the same group(P<0.05).However,compared the concentration between on the first and on third day,there was no statistic significance(P>0.05).(5)The concentration of SDF-1αon the 1st day of AMI group were obviously higher than those on the 3rd and 7th day of the same group(P<0.05).However,compared the concentration between on the 3rd day and on the 7th day,there was no statistic significance(P>0.05).(6)Obvious positive correlation was noted either between CD34+BMSC and VEGF(r=0.583,P<0.05)or between CD34+BMSC and SDF-1α(r=0.628,P<0.05)in AMI group;Obvious positive correlation was noted between CD34+BMSC and LVEF(r=0.637,P<0.05);Obvious positive correlation was noted either between VEGF and CK(r=0.791,P<0.001)or between VEGF and CK-MB(r=0.833,P<0.001),too.Conclusion:The bone marrow-derived stem cells(BMSC)were obviously increased in the peripheral blood of AMI patients,and shew positive correlation with VEGF and SDF-1α.So that it is inferred that when the organism had suffered from AMI the dynamic change of VEGF and SDF-1αcould be contribute to the mobilization and homing of BMSC to repair the injured myocardium.
