Objective The aim of this study was to investigate the effect of plasma concentrations of transforming growth factor (TGF)-betal (TGF-) and plasminogen activator inhibitor type 1 (PAI-1) on diabetic atherosclerosis.Method Color Doppler ultrasound was used to measure the intima-media thickness (IMT) and plaque of the common carotid artery in 62 cases of type 2 diabetic mellitus (T2DM). The patients were assigned to three groups based on IMT: the group with normal IMT (IMT<0.8mm); the group with thicker IMT (0.8mm lmm ). Fasting plasma TGF and PAI-1 concentrations were measured by ELISA in 62 cases of type 2 DM and 28 age- and sex-matched health controls.Results (1) By analysis of variance there are no significantly difference of sex, age, course of diseases, body weight index (BMI), cholesterol (TC), glycerol(TG), low-density lipoprotein(LDL) and fasting blood glucose(FBG) in those groups of T2DM. (2)The plasma concentrations of TGF and PAI-1 in those groups of T2DM were significantly higher than those in health controls (p<0.01) ; The plasma concentrations of TGF- and PAI-1 in the group with thicker IMT were significantly higher than those in the group with normal IMT (p<0.05 ) ; the plasma concentrations of TGF in the group with plaque were significantly lower than those in diabetes with thicker IMT (p<0.05 ) the plasma concentrations of PAI-1 in the group with plaque were significantly higher than those in diabetes with thicker IMT (p<0.05 ) . (3) Compared tothe control group, the patients with high plasma concentrations of PAI-1 were increased in those groups of T2DM (p<0.05). (4) Bivariste correlative analysis show, The plasma concentrations of PAI-1 in T2DM were positively correlated with BMI, HOMA-IR, TGF and IMT. (5) Bivariste correlative analysis show, the IMT was significantly correlated with PAI-1 , HbA1c, SBP, DBP and HOMA-IR. (6) By stepwise regression analysis, PAI-1 and SBP were the independent factors for IMT in T2DM.Conclusion The change of plasma TGF and PAI-1concentrations followed by diabetes Mellitus play pivotal roles in the pathogenesis and development of atherosclerosis.
