Objective: To investigate the cure effects and mechanism of Qing Guang An Granule(QGAG) against the injury on retina in rabbit Eyes with acute ocular hypertension.Methods: 49 rabbits were divided randomly into seven groups and there were seven rabbits in every group. A\ and A2 groups were treated with the solution of QGAG in stomach; BI and B2 groups with the solution of Shi Hu Ye Guang Pill(SHYGP);Ci and C2 groups were regarded as the model control groups; the D group was regarded as the blank control group. After the rabbits had been fed and administrated for one week, the acute ocular hypertension models were established by perfusing physiological saline in the anterior. The activities of Adenosine Triphos Phatase(ATPase) , Sucdnate DeHydrogenase (SDH) and Glutathione Peroxidage (GSH-PX) in retina tissue were measured in one week and two weeks respectively after the treatment. At the same time, we observed thepathologic changes of retina tissue and the apoptosis by light microscopy and transmission electron microscope.Results: The activities of ATPase, SDH and GSH-Px in retina tissue were significantly higher in the QGAG group than those in SHYGP group and the model group at a time(P<0.01). After two weeks, the activities of san enzymes were drew near those in blank control group. However, there was significant difference(P<0.05). Compared with first week, the pathologic changes of retina tissues were little at second week Light microscopy showed the retinal structure was in disorder, the contents of visual cells decreased to a greater or lesser degree in the model control groups and the SHYGP groups and the apoptosis were higher than those in the QGAG group(P<0.01). Electron microscope showed most ganglion cells were edematous, the mitochondria were swollen, the rough endoplasmic reticula were dilated and apoptosis appeared. However, these changes were little in the QGAG group.Conclusions: l.QGAG could evidently increased the activities of ATPase , SDH and GSH-Px. 2.QGAG had protective effects on retina. The mechanism was related to recovering normal energy metabolism and elevating the ability of anti-oxidation in retina. 3.Apoptosis appeared in the outer nuclear layer, the inner nuclear layer and RGC of glaucoma retina.
