Elsholtzia blanda is the total herb of Elsholtzia blanda (Benth.)Benth., which is abundant in tropical area of southwest China. According to the folk residents of Hani and Lahu ethnic minorities, its inflorescence part could significantly relieve Xiongbi symptom which in Traditional Chinese Medicine refers to the syndromes of coronary heart disease. But it has not been included into national drug standard or any regional medicinal materials criterion. Given the fact that little is known about effectively preventing and treating those common killing diseases so far, and the few existant theraputical drugs have many drawbacks in the safety and effectiveness aspect[1], it is still the arduous tasks for researchers in pharmaceutial sciences to explore new anti-Xiongbi symptom drugs with exclusive intellectual property rights of China. Since its major effective component is total flavone extracted from inflorescence of Elsholtzia blanda[2], we adopted the modern techniques in experimental pharmacological science to explore the effects and mechanisms of total flavone of Elsholtzia blanda(TFEB) on rats' Xiongbi symptom, as well as its antiarrhythmia effect induced by myocardial ischemia or other factors on rat models. Thus we provide tangible evidence for the development of TFEB as a novel herbal Chinese medicine treating Xiongbi symptom.1. Protective effects and mechanisms of TFEB on myocardial ischemia in rats 1.1 Effect of TFEB on myocardial ischemia by left anterior descending coronary artery (LAD) ligation in ratsMale rats were given TFEB by ig for 5 d, then acute myocardial ischemia model was prepared by LAD ligation. After 3 h, the rats were decollated and the leftventricle was dyed by nitrotetrazolium blue chloride(NBT) to distinguish infarction area from non-inffarction area. Myocardial infarction index was calculated accordingly to assess the effect of TFEB on myocardial ischemia. A piece of ventricular muscle come from apex of heart was dyed by hematoxylin and eosin(HE) to observe the degree of myocardial lesions by microscopic examination. Nitric oxide (NO) level in serum was measured to demonstrate the relevant mechanism.The results indicated that TFEB could markedly reduce the myocardial infarction index in a dose-dependent fashion (P<0.05-0.01). Microscopic examination showed that the myocardium of sham group had no lesion in morphology and the muscle fiber kept integrity and the cardiac myocyte lined up in order. While control group displayed extensive swelling, rupture, confluent, even break in cardiac myocyte. TFEB could relieve the lesion degree of myocardium. NO level in serum after LAD ligation in TFEB 100 and 150 mg/kg groups was markedly reduced (P<0.05-0.01). The results suggest that TFEB has protective effect on myocardial ischemia induced by LAD ligation in rats and can prevent Xiongbi symptom, possibly via the regulation of NO level in serum.1.2 Effect and mechanism of TFEB on myocardial ischemia induced by pituitrinum in ratsMale rats were given TFEB by ig for 5 d, acute myocardial ischemia model was produced by iv pituitrinum (pit) . Degree of ischmia was assessed by T amplitude of lead II dynamic ECG and heart rate. The mechanism of anti-myocardial ischemia was investigated by the level of creative phosphokinase(CK), calcium(Ca2+), NO in serum and malondialdehyde(MDA), Ca2+, NO in myocardial homogenate. The results indicated that at the dosage of 200 mg/kg, TFEB could markedly reverse the elevated T amplitude of ECG and slow heart rate induced by pit (P<0.05) .The serum level of CK and MDA were significantly decreased. TFEB could decreased myocardial Ca2+, increse Ca2+ in serum ( P<0.05~0.01 ) and relieve Ca2+ overload of ischemic myocardium. The level of NO in serum and myocardial tissue were significantly increased in TFEB groups(P<0.05~0.01). The results suggest that the protective effect of TFEB to myocardial ischemia in rats is...
