Dandouchi, also named Xiangchi.Danchi, as a traditional Chinese medicine, is first recorded in the ancient Chinese medical work , made by fermented soya beans, and is a main component of popular prescription Yinqiao Jiedu Pills . It has the effects of inducing diaphoresis (Jiebiao), relieving restlessness(Chufan), and dispelling emission heat (Xuanfayure). It is used to treat flu, headache, fidgeting due to deficiency and insomnia (Xufanbumian), etc.Dandouchi contains various kinds of active principles such as soy isoflavones, soy Saponins, vitamins, amino acids, enzymes, etc. It has been found that Soy isoflavones have both weakestrogenic and weak anti-estrogenic effects, and may also have the antioxidant, antidipsotropic, anticarcinogenic, anti-atherogeru'c and anti-osteoporotic activities, and have been used to treat tumors, osteoporosis, and so on. Soy saponins have the effects of adding taste, acting on viruses, fungi, increasing immune system strength, reducing cancer risk, lowering cholesterol, etc. 1 Technique in manufacture and extractionThe various condition in procedures of manufacture were studied. The optimum manufacture technology was as followed: the selected soybean were immerged in soup decocted by southernwood and mulberry for 6h, and then steamed for ISO minutes under normal pressure, and then fermenting for 7 days mixed with finished product of Dandouchi, and then dried for next process.Extraction and purification of isoflavones from Dandouchi powder were studied. By a single factor and a orthogonal test, with the extraction efficiency of isoflavones as study targets, the influences of extraction solvent, sample and extracting ratio, extracting time, times of extraction were studied. The optimum extraction technology was as followed: solvent 70%ethanol, solvent to Dandouchi weight at 1:3, three times extraction, and extracting time is 2h.The enrichment and purification of isoflavones from ethanol extraction had been performed through macroporous resin column. To study the technological parameters of the enrichment process, with the enriched degree of daidzein as a marker, the type of macroporous resin, the absorbing temperature, pH value and eluting solvent were studied. The optimum technological parameters are as followed: temperature at 15, pH value at 5.0, the 905 type of macroporous resin were selected to the enrichment and purification process and 50% ethanol were selected as eluting solvent. Under the condition, the daidzein of 8% purity could be obtained by separated through macroporous resin column.The application of Near-infrared spectroscopy(NIRS) in quality control in active principles extracting of DandouchiNIRS would be occurred when there is absorption due to overtone and combination bands originating from fundamental molecules vibrations correspond mainly to C-H N-H and 0-H groups in compounds. It has advantages over non-destructive, non-invasive, reagent-free, no sample preparation, on-line method. Various kinds of soy .fermented sauces and Dandouchi could be distinguished by cluster analysis.The functional components of Dandouchi wererapidly by NIRS technology. The optimum condition of mathematics model two components (daidzein, genistein) was studied, including the sample set selection, chemical value analysis, the detection methods and conditions. The analytical results demonstrated that the correlation between the chemical value (by HPLC) of the two components of sample set and the NIR predicated value is 0.993 for daidzein and 0.990 for genistein. The correlation between the chemical value of test sample set and the NIR predicated value is 0.981 and 0.965, respectively. It proved that the mathematical model that we establish could be used in practice.2 The prevention and therapy of "Xufanbumian" by Dandouchi extraction in ratsIt has been demonstrated that Isoflavones have a significant ability to prevent osteoporosis in both animal models and humans due to its weak estrogen-like activity.
