Determination, Extraction And Purification Of. Momordica Total Saponins

Semen Momordicae,the dried and ripe seed of Momordica cochinchinesis Spreng(cucurbitaceae)is a traditional Chinese medicine considered to exhitbit a cooling and resolvent effect,commonly used as a remedy for wounds,bruises,swelling,hemonhoids and pus.Saponins are the major effective components in the medicine of Momordica cochinchinesis Spreng.A determination method of total saponins in Momordica cochinchinesis Spreng,the optimal parameter of extracting the total saponins by ultrasonic and the purification technology by AB-8 resin have been studied in this paper.The result shows:1.A determination method of total saponins in Momordica cochinchinesis Spreng was established by measuring using chromatometry at 538nm.The calibration curve was linear in range of 3.92-23.5mg/L(r =0.999)and the range of average recovery were 97.93%,RSD=2.02%.The method was simple and effective.2.The optimal parameters of the total saponin extraction from Momordica cochinchinesis Spreng by ultrasonic are as following:the concentration of alcohol 50%,material/solvent ratio 1:12,extracting 30min and tempreture 40℃.3.AB-8 macroporous resin shows better performance for purifying the Momordica cochinchinesis Spreng total saponin,and its static adsorption ratio is 41.87mg/g dried resin,the elution ratio is 85.49%;The parameters of dynamic adsorption are as fowllowing:the original concentration 0.99mg/mL,the feeding rate 1BV/h and pH value 3.0;The parameters of dynamic elution are as fowllowing:firstly eluting the water-soluble impurity like amylose by distilled water,then eluting by 30%alcohol, lastly eluting by 50%alcohol,collecting the part eluted by 50% alcohol;the total saponin in the solid after purification is more than that in the solid before purification,the refinement ratio can reach 1070%which shows the effect that AB-8 macroporous resin purifies the Momordica cochinchinesis Spreng total saponin is well.
