Objective: To explore the variation of the proteins correlated with chemo resistance of the human lung cancer cell lines with the treatment by anticancer drugs in different concentration.Methods: The human lung cancer cell lines (LPET-a-2 and SPC-A-1) were treated by anticancer drugs-doxorubicin, etoposide, cisplatin and combination of the three drugs with different concentrations. After being treated, MDR1, MRP, LRP, ref-1, NF-KappaB, MDM2, GST, TS, MT and P27 in the human lung cancer cell lines were tested by flow cytometry. The percentage of cells expressing these proteins and the mean quantity of them were acquired by the fluorescence quantity analysis. We could calculate the number of the cells expressing each protein, the mean quantity of each protein in each cell and the total one in each test group after each treatment by different drugs with different concentration.Results: 1. The expression of these proteins in SPC-A-1 cell line was lower than that in LTEP-a-2 cell line, and its concentrations of IC50 of the three drugs were smaller than those in LTEP-a-2 cell line.2. The number of the cells expressing MDR1 in ADM groups of different concentrations was close to that of control and its mean quantity of MDR1 was higher than that of control after treatment. The number of the cells expressing MDR1 in the other three drugs' groups and its mean quantity of MDR1 increased as the more drugs were added. The total quantity of MDR1 in ADM's groups of different concentrations was 2 to 5 times higherthan that in control, but it was almost the same among the ADM's groups with different concentration. The total quantity of MDR1 in the other three drugs' groups increased as the more drugs were added.3.The expression of MDR1, LRP, ref-1, NF-KappaB, MDM2, GST, TS, MT and P27 incresed as the more drugs were added in some groups in LTEP-a-2 cell line, but there are not the rules in other groups. The expression of MRP, NF-KappaB and MT incresed as the more drugs from IC20 to IC40 were added in some groups in SPC-A-1 cell line.Conclusion: The expression of the proteins related to chemo resistance is associated with the degree of the cell's resistance to anticancer agents. The mechanism inducing chemo resistance of the different cells was not the same. The expression of different proteins was induced by the same anticancer agents in different cell lines, while The expression of different proteins was induced by different anticancer agents or different concentration of the same drugs in the same cell line. The combined chemotherapy was more effective than the single-agent one. These data suggest that the patients suffering from cancer should be given individualized and combined chemotherapy for treatment.
