Objective: to investigate differential regulation of unprimed- and primed- lymphocyte activation by E2.Methods: the proliferation mouse lymphocyte activation was measured with MTT assay, antibody activity of supernatants was detected by indirect ELISA, and TGF-P level in supernatants was assayed by indirect ELISA.Results: E2, ranged from 60pg/ml to l00ng/ml, was found to increase proliferation of mouse lymphocytes, especially, of primed lymphocytes. E2 at lower concentrations inhibited antibody production. Conversely, E2 promoted antibody production at a higher concentration. TGF-p was undetectable in the supernatants except the ones from primed lymphocyte culture in the presence of E2 at 50 or lOOng/ml and ConA at 0.6ug/ml.Conclusions: E2 preferentially regulates primed lymphocyte activation. It might not regulate the responsiveness of naive lymphocytes to antigenic stimulation.
