A Study on the Expression of Bcl-2 and Proliferating cellnuclear antigen in Postmenopausal Leiomyoma Background:Uterine leiomyoma are the most common benign tumor in femal genital tract. Steroids hormone are promoters of leiomyoma growth .Traditionally,the majority of them regress after menopause,but some of them have no reduction in tumor size,and even cause symptoms.Recently,investigators have suggested that cell proliferation and apoptosis maybe important in regulation of leiomyoma growth.The aims of this study were to investigate the expression of Bcl-2 and PCNA in premenopausal and postmenopausal leiomyoma.The expression of Bcl-2 and PCNA in 117 cases were examined by immunohistochemical technique,including 40 cases premenopausal leiomyoma,77 cases postmenopausal, 57 cases with symptomatic postmenopausal leiomyoma,20 cases with regressive postmenopausal leiomyomas,20 cases of proliferative phase,and 20 cases of secretory phase of menstrual cycle,named as group A, B, C, D, E, and F respectively.The relationships between expression of Bcl-2 and PCNAwere analyzed.The results as follow:1. The expression of Bcl-2The expression level of Bcl-2 in premenopausal leiomyoma was significantly lower than that of postmenopausal leiomyoma(p<0.05).The expression level of Bcl-2 in symptomatic postmenopausal leiomyoma was significantly higher than that in regressive postmenopausal leiomyoma(p<0.05);The expression level Bcl-2 in secretory leiomyoma was higher than that in proliferative leiomyoma.The expression level of Bcl-2 in leiomyoma was significantly higher than that in the corresponding myometrium in premenopausal leiomyoma and symptomatic postmenopausal leiomyoma(p<0.05),but no difference was shown in regressive postmenopausal leiomyoma.2. The expression of PCNAThe expression level of PCNA appeared a trend to decreace in leiomyoma from premenopause to postmenopause.The expression of PCNA in premenopausal leiomyomas was significantly higher than that in postmenopausal leiomyoma(p<0.05).The expression of PCNA in secretory leiomyoma was higher than that in proliferative leiomyoma(p<0.05).The expression level of PCNA in leiomyoma wassignificantly higher than that in corresponding myometrium inpremenopausal leiomyoma and symptomatic postmenopausaslleiomyoma(p<0.05),but no difference was shown in regressivepostmenopausal leiomyoma.The expression level of PCNA insymptomatic postmenopausal leiomyoma was significantly higher thanthat in regressive postmenopausal leiomyoma(p<0.05).3. The relationship between the expression of Bcl-2 and PCNAThe expression of Bcl-2 was no correlated with that of PCNA inleiomyoma of all groups.4.The relationship between mitotic counts per 10 high power fields andBcl-2,PCNA.Mitotic counts per 10 high power fields in postmenopausal leiomyomawere significantly lower than those of premenopausalleiomyoma(p<0.05).The growth activity of leiomyoma was effected bysteroids level.Mitotic counts per 10 high power fields in symptomaticpostmenopausal leiomyoma were significantly higher than those inregressive postmenopausal leiomyoma(p<0.05).Proliferative activity insymptomatic postmenopausal leiomyoma was higher.Mitotic countswere positively and significantly correlated with the expression level ofPCNA in leiomyoma,while there were no correlation in normal myometrium.Mitotic counts were no correlated with the expression level of Bcl-2 in leiomyoma and normal myometrium.The results indicated that Bcl-2 and PCNA could increase the growth of leiomyoma by different way.
