Colorectal carcinoma(CRC) is one of the most prevelant malignancies in gastro-intestinal oncology.Its invasion and metastasis play an important role in prognosis.To research the relationship of fibronectin(FN) , integrin a v , integrin|33 and metastasis of colorectal carcinoma,30 cases of colorectal tumors were collected to detect the expression of fibronectin in extracellular matrix (ECM) through reverse transcription polymerase chain reaction(RT-PCR) and immunohistochemistry. At the same time the expression of integrin subunits a v, (33 among cellular membrane was assayed by RT-PCR.The results show that:1. The mRNA transcription and protein expression levels of fibronectin in CRC are significantly higher than those in tissue surrounding tumor (P2,P5) and tissue of cutting side (N).2. There is significantly positive correlation between the mRNA transcription and protein expression levels of fibronectin in pericellular interstitial of colorectal carcinoma and neoplasia invasion,lymph node metastasis as well as Dukes stage.The fibronectin expression appeares significantly higher in invasion subgroup compared with that in non-invasion subgroup.3. The fibronectin expression levels in pericellular interstitial of CRC are neither found correlated with CRC histological types nor companying with liver metastasis and differertiation status.4. The deletion of FN in CRC basement membrane is found significantly positive correlated with CRC neoplasia invasion,lymph node metastasis and Dukes stage.The fibronectin expression in CRC basement membrane appears significantly higher in invasion subgroup compared with that in non-invasion subgroup.5. There is significantly negative correlation between FN expression levels and liver metastasis.However no significant difference is found in expression levels.6. No correlation was found between the deletion of FN in CRC basement membrane and CRC histological types anddifferentiation status.The mRNA transcription expression levels of integrin a v and integrin P3 in CRC are sifnificantly higher than that in tissue surrounding tumors(P2,P5) and tissue of cutting side (N). Conclusions:. The high expression of fibronectin in CRC pericellular interstital may increase the potency of CRC invasion and metastasis.The significant deletion of FN in CRC basement membrane may suggest that CRC possess much more potency of invasion and metastasis.The significantly higher mRNA expression levels of integrin a v and integrin p3 may lead to increasing the activation of fibronectin-integrin a vp3-cytoskeletal protein system,which increase the potency of colorectal carcinoma invasion and metastasis, however,the hypothesis above need to be researched further.. The high expression of FN in colorectal carcinoma pericellular interstital and low expression of FN in CRC basement membrane is possibly be regarded as an index screening CRC subgroup of strongly potential invasion and high frequent metastasis.To test expression of FN may be helpful to evaluate colorectal carcinoma patient prognosis.
