Objective: To study the expressions and quantitative change of collagen components, such as collagen type I and collegen type IV , in glomerular areas of mesangial Proliferative Nephritis (MsPGN) in adults, to identify the correlations between the quantitative changes of collagen components and the degrees of pathological changes. Methods: Collagen type I and collagen type IV in glomerular areas was detected by immunohistochemical staining in total 56 samples of mild ,media and grave MsPGN , and normal kidney tissues, which was attained from the ablated kidney of patients with renal cancor. We applied immunohistochemical staining assay (streptavidin-peroxidase,SP) to study the expressions of collagen type I and collagen type IV, used image analysis system (HPIAS-1000) to analysis the stained surface areas of the two collagen components. Results: There is no expression of collagen type I in normal glomerular areas while mild expressions of the same components exist in renal tubular interstitial areas. There are different expressions of collagen type I in glomerular areas of MsPGN. There are positive expressions of collagen type IV in glomerular and tubular interstitial areas either of MsPGN or of normal kidney tissue. In glomerular areas with fibrosis, there arc intense positive expressions of collagen type I and collagen type IV. The two component expression difference between grave MsPGN and mild or media MsPGN have significance(P< 0.05), and the difference between expressions of collagen type I or collagen type IV in mild and media mesangial proliferative glomerular areas also have significance (P<0.05). We also find that the two collagens distributed intensively and extensively in sclerotic glomeruli, and that the expressions of collagen type I and collagen type IV have positive correlation, whose coefficient was 0.628. Conclusion: This study have demonstrated that the expressions of collagen type I and collagen type IV increase in glomerular areas with the pathological changes getting intense, the two collagen expression tensity is different in grave ,media or mild mesangial proliferative glomerular areas. Therefore, the collagen component analysis can be used as one of the standards that can indicate the degree of pathological change in MsPGN, may help nephrologists or physicists make properly therapeutic plans and provide them the evidence of evaluating the prognosis of MsPGN.
