Purpose the purpose of study is to evaluate mechanismwhich antisense sequence of thrombin receptor influence VascularSmooth Muscle Cell Proliferation.Mater i a I and methods Ce 11 transf ect i on, RT-PCR, West-B I ot, 3H-TDRunincorporated assay, 3H-inositol unincorporated assay, EILSAassayare used to measure Vascular Smooth Muscle Cell Proliferation andexpression of thrombin receptor of Vascular Smooth MuscleCel I in order to detect mechanism of antisense sequence of thrombinreceptor which influence Proliferation of Vascular Smooth MuscleCell.Result Antisense sequence of thrombin receptor clearly restrainpro I i f erat i on of Vascu I ar Smooth Muse I e Ce 11 and mRNA and prote i nexpression of thrombin receptor and metabolism of 3H-inositol .Therewere greatly statistic difference among them.Conclusion Antisense sequence of thrombin receptor restrained SDrat vascular smooth muscle cells proliferation marketedly due torestrain gene expression of thrombin receptor and signal induction .Thrombin receptor has important role in Proliferation of VascularSmooth Muscle Ce11. Thus, it provide basis of theory and experimentwhich antisense technology may cure vasculardisease(atherosclerotic and restenosis after percutaneoustransluminal coronary angioplasty) and establish fundament whichfurther constructed antisense expression carrier treat vasculardisease.
