Objective: To develop a rat model of insulin resistance,and to study the effect of kakonein injection on expression of protein kinase B in insulin resistance rats.Methods: 30 female SD rats were randomly divded into two groups. The model group were given the high fat diets(30%pig fat, 10%sucrose, 3%cholesterol) for 8 weeks. Then 9 rats of the group have been choosed as the kakonein treating group and were given abdominal injection (100mg kg-1 d-1). After 4 weeks of treatment, plasma glucose, plasma insulin, as well as skeletal muscles pathology and visceral fat were observed. Western blotting and computer image pattern analysis system were used to analysis expression of PKB in skeletal muscles.Result: (1)The model group showed a hyperglycemia, hyperinsulinism and obviously obesity by high fat-feeding, the insulin resistance index (ISI) decreased while the HOMA insulin resistance index (HOMA-IR) increased compared to the normal group, insulin resistance was induced in this way. (2) Compared with the pathology control grouprfasting blood glucose (FBG), fasting insulin (FINS), Leelndex, visceral fat mass and the HOMA insulin resistance index (HOMA-IR) of the treating group were significantly decreased whereas the insulin index (ISI) enhanced. The ultrastructure of skeletal muscle recovered near normal. Expression of PKB was markedly increased by kakonein treating.Conclusion: (1) High fat diets can successfully induce insulin resistance model. (2) Kakonein injection can significantly elevate exepression of PKB and ameliorate the state of insulin resistance. Improving biologic effect of insulin and preventing the deleterious effect of fat may also be involved in the mechanism concered.
